摘　　要：目的通过研究维生素D受体(VDR)在妊娠期肝内胆汁淤积症(intrahepatic cholestasis of pregancy,ICP)孕妇外周血、脐血及胎盘中的表达,探讨VDR与ICP发病的关系。方法采用荧光定量PCR法检测ICP组和正常对照组外周血、脐血及胎盘组织中的VDR的mRNA表达,ICP组和正常对照组均为52例。两组孕妇胎盘中VDR蛋白表达水平采用免疫组化法检测。结果 (1)ICP组外周血、脐血及胎盘组织中VDR的mRNA表达水平均明显低于对照组,差异有统计学意义(P值均<0.05),ICP组中VDR mRNA在脐血中的表达明显高于在外周血及胎盘中的表达,差异有统计学意义(P值均<0.05);(2)ICP组胎盘中VDR的蛋白阳性表达率明显低于正常对照组,差异有统计学意义(P<0.05)。结论 VDR在ICP孕妇外周血、脐血及胎盘中的表达均低于正常孕妇,可能在ICP发病中起一定的作用。Objective： To investigate the expression of the vitamin D receptor （VDR） in the peripheral blood, umbilical cord blood and placenta tissue in patients with intrahepatic cholestasis of pregnancy （ICP） , and to discuss their relationship between VDR and the pathogenesis of ICP.Methods： The expression of VDR at mRNA levels in the peripheral blood, umbilical cord blood and placenta tissue were assayed by using real time fluorescent quantitative reverse transcriptase polymerase chain reaction （real-time qRT-PCR） in the two groups.ICP group were 52 pregnant women, and 52 normal pregnant women in the normal group.The expression of VDR at protein levels were assayed by using SP immunohistochemical method in the two groups. Results： （ 1 ） The expression of VDR at mRNA levels in the peripheral blood, umbilical cord blood and placenta tissue in ICP group were obviously lower than in normal group （P〈0.05） , The expression of VDR levels in the umbilical cord blood was significantly higher than in the peripheral blood and in placenta tissue. （2） The expression of VDR at protein levels in the placenta tissue was significantly higher in normal group than in ICP group （P〈0.05） .Conclusions： Low VDR expression in the peripheral blood, umbilical cord blood and placenta tissue of patients with intrahepatic cholestasis of pregnancy may play an important role in pathogenesis of ICP.
