摘  要:目的:利用GAL4/UAS基因表达调控系统研究CG9940基因过表达对果蝇心脏抗衰老功能、运动能力和寿命的影响。方法:W1118雌性果蝇与arm-Gal4雄性果蝇杂交取F1为对照组,y[1]w[67c23]P{y[+m8]=Mae-UAS.6.11}CG9940[GG01267]雌性果蝇与arm-Gal4雄性果蝇杂交取F1为过表达组,2组均收集处女蝇1 200只,在第1、3、5和7周进行检测。实时荧光定量PCR测定表达量,M-mode心动图测量心管舒张功能不全指数、心率失常指数,测量攀爬指数和统计生命周期。结果:平均相对表达量过表达组高于对照组(P<0.01),过表达率为144.09%;舒张不齐指数在第7周对照组高于过表达组(P<0.01),对照组舒张不齐指数第5周到第7周有非常明显上升(P<0.01);心率失常指数对照组第1周、第5周和第7周均高于过表达组(P<0.05),对照组心率失常指数第3周到第5周有明显上升(P<0.05);第1周和第3周龄果蝇攀爬指数过表达组强于对照组(P<0.01);过表达组果蝇平均寿命长于对照组(P<0.01),平均延寿率为20.4%,对照组的最高寿命是64 d龄,过表达组的最高寿命是77 d龄。结论:CG9940基因过表达能够有效保护中老龄、老龄果蝇心脏功能,提高幼龄和中龄果蝇运动能力,有助于老龄果蝇活动能力的维持,具有延长寿命的作用,提示CG9940基因与中老龄心脏功能增龄衰退、运动能力及寿命有着重要联系。Objective: This study explored the effect of over-expression of CG9940 on anti-aging cardiac function, exercise ability and lifespan in drosophila melanogaster by using Gal4/UAS systenL Methods: The control group drosophila melanogasters are the filial generation of IF-111s female drosophila melanogasters crossing with arm-Gal4 males, and the over-expression group ones are the filial generation of yE[1] wE67c23] P{ Y[ + m81 = Mae-UAS. 6.11 } CG9940[GG01267]females crossing with arm-Gal4 males. One thousand and two hundreds virgin drosophila melano- gasters in each group were tested when they were one-week, three-week, 5 - week and seventh-week old. The mRNA expression of CG9940 gene was detected by real-time PCR. The diastolic dysfunction index and arrlwthmia index were detected by M-mode eehoeardiography, and climbing index and lifespan were calculated. Results : The mRNA expression of CG9940 gene was higher in over-expression group than in control group (P 〈 0.01 ) and the over-expression rate is 144.09%. Diastolic dysfunction index of seven-week old drosophila melanogasters in con- trol group is higher than that in over-expression group ( P 〈 0.01 ) , and it increased significantly from five-week old to seven-week old in control group (P 〈 0.01 ). Arrhythmia index of one-week, five-week and seven-week old ones in control group is higher than that in over-expression group respectively ( P 〈 0.05 ) , and it increased signif- icantly from three-week old to five-week old in control group ( P 〈 0.05 ). The climbing index of one-week old and three-week old drosophila melanogaster in control group is lower than that in over-expression group (P 〈 0.01 ).The average lifespan in over-expression group is 20.4% longer than that in control group (P 〈0.01 ) ; the oldest one in over-expression group is 77 day-s old while it is 66 day-s old in control group.
