摘 要:通过液体培养的方法研究了解磷枯草芽孢杆菌HL-1在培养168 h内溶磷能力、活菌数及培养介质pH值的动态变化,并分别以培养基初始pH值、碳氮源种类及最佳碳源浓度为变量,设置单因素试验,探究不同培养条件对供试菌株溶磷能力的影响。结果表明:HL-1的溶磷能力与培养液的pH值之间呈极显著的负相关,其初始pH值为5.0,以10.0 g/L的葡萄糖为碳源,硫酸铵为氮源的培养条件下显示出最高的溶磷活性,培养168 h后的溶磷量为105.82 mg/L。试验结果反映出微生物的溶磷能力受到培养环境中诸多因子的影响,优化各个因子的组合,可以在实验室条件下提高微生物的解磷效果,同时可为生物菌剂制备工艺的开发提供理论参考。A liquid culture experiment was conducted to study the dynamic variations of the phosphate-solubilizing capability and growth of the tested Bacillus subtilis HL-1 and medium pH within 168 h as well as the different effects of initial pH of culture media, carbon sources, nitrogen sources and the level of carbon concentration on phosphate- solubilizing capability of HL-1. The results indicated that a significant negative correlation was observed between the phosphate-solubilizing capability of HL-1 and the medium pH. The HL-1 displayed the best phosphate-solubilizing capability on condition that the initial pH of medium was 5.0, glucose with 10.0 g/L for carbon source and ammonium sulfate for nitrogen source. The soluble phosphorus content achieved 105.82 mg/L after the strain cultured for 168 h. The test results reflect that the microbial phosphate-solubilizing capability can be influenced by many factors of the culture environment, the optimal combination of factors will not only be conducive to its effect under laboratory conditions, but also provide a theoretical basis for the development of microbial preparation process.
