摘 要:建立了用纳氏比色分光光度法同时快速测定土壤中速效氮和总氮含量的方法。对土壤中速效氮的提取方式、提取液、提取时间等条件进行了优化,选用20%的氯化钠溶液超声1小时提取土壤中的速效氮。土壤样品用硫酸双氧水加氢氟酸微波密闭消解半个小时,消解液和速效氮提取液用纳氏比色法测定。方法成功用于土壤样品中速效氮和消解氮的测定,回收率为86%~102%,对国家标准物质GBW07413a(ASA -2a)进行测定,测定值均在推荐值的误差范围内。该法设备操作简便,适用于土壤中速效氮和总氮的同时测定。A method was established for the simultaneous determination of available nitrogen and total nitrogen in soil by Nessler's colorimetry spectrophotometry. The extraction conditions such as extraction equipment ,the available nitrogen extractant ,and the extraction time were optimized ,with 20% sodium chloride by one hour to extract available nitrogen in soil. Soil samples was digested by sulfuric acid-hydro-gen peroxide-hydrofluoric acid with microwave for half an hour ,the digeted liquid and the available extract was detected by spectrophotometer after Nessler's colorimetry. The developed method was applied suc-cessfully to the detection of available nitrogen and total nitrogen in soil samples ,and the recovery rate was 86% -102%. The national standard substance GBW07413a (ASA-2a) was also measured ,the measured value was in the range of permitted prror. The method is simple ,rapid and accurate ,and it is appropriate for the simultaneous determination of available nitrogen and total nitrogen in soil.
