摘 要:目的:研究补肾益气方药对自然衰老大鼠CD4+T细胞核转绿因子κB(NF-κB)核转位与白细胞介素2(IL-2)基因转录的影响。方法:以自然衰老大鼠为模型,分组:青年对照组(3月龄);老年对照组(20月龄);老年补肾益气组(20月龄)。老年补肾益气组饲喂补肾益气方药,其余两组给予正常饮水,3个月后,流式细胞仪分选CD4+T细胞,以anti-CD3/anti-CD28进行刺激。Real time q PCR法检测IL-2 mRNA的表达;ELISA法检测血清IL-2蛋白浓度;激光共聚焦检测NF-κB细胞内定位;Western blot法检测NF-κB胞内表达。结果:与青年对照组比较,刺激后老年对照组CD4+T细胞IL-2 mRNA表达显著降低(P<0.01);刺激0.5h时,胞浆NF-κB表达明显增高,核内NF-κB表达明显降低(P<0.01)。与老年对照组比较,老年补肾益气组CD4+T细胞IL-2 mRNA表达及血清浓度均显著升高(P<0.01);刺激0.5h时,胞浆NF-κB表达明显降低,核内NF-κB表达明显升高(P<0.05)。结论:老年大鼠CD4+T细胞对anti-CD3/antiCD28刺激的敏感性下降,胞内NF-κB核转位能力减弱。补肾益气方能通过活化衰老CD4+T细胞NF-κB的核转位,增强NF-κB与IL-2启动子的结合,提高IL-2表达,延缓衰老细胞免疫功能衰退。Objective: To study the effects of reinforcing kidney and nourishing qi on the nuclear translocation of nuclear transcription factors kappa B (NF-κB) and the gene transcription of interleukin-2 (IL-2) in the aged rats CD4+T ceils. Methods: The nature old rats were used as the senile animal model, the 3 months old rats as the young control group, the 23 months old rats as the old control group, and the 20 months old rats were treated by reinforcing kidney and nourishing qi recipe as the treatment group. The CD4+T cells stimulated by anti-CD3/anti-CD28 were detected with flow cytometer. The expression of IL-2 mRNA was detected with Q RT-PCR. The expression of IL-2 protein in serum was detected with ELISA. The location of NF-κB was detected with confocal laser scanning microscope. The expression of NF-κB in cell's nucleus was detected with Western blotting. Results: The expression of IL-2 mRNA of old control group in CD4+ cells was significantly lower than that of the young control group (P〈0.01). After 0.5 hour, the expression of intracytoplasmic NF-κB of old control group was significantly higher than that of the young control group, and the expression of NF-κB in cell's nucleus was significantly lower (P〈0.01). The expressions of IL-2 mRNA of treatment group in CD4T cells and in serum were significantly higher than those of old control group (P〈0.01). After 0.5 hour, the expression of intracytoplasmic NF-κB of treatment group was significantly lower than that of old control group, and the expression of NF-κB in cell's nucleus was significantly higher (P〈0.05). Conclusion: The sensitivity of CD4+T cells stimulated by anti-CD3/anti-CD28 was reducing and the nuclear translocation of intracytoplasmic NF-κB was weakened. The reinforcing kidney and nourishing qi recipe could activate the nuclear translocation of NF-κB in senile CD4+T ceils, enhance the combination of NF-κB and promoter of IL-2 and increase the expression of IL-2 that could delay the dete
