益气解毒活络方对早期糖尿病肾病大鼠肾组织结缔组织生长因子及Ⅳ型胶原mRNA的影响

摘　　要：目的:研究益气解毒活络方对链尿佐菌素(STZ)诱导的糖尿病肾病(DN)大鼠肾脏结缔组织生长因子(CTGF)及Ⅳ型胶原(ColⅣ)mRNA的影响。方法:Wistar大鼠90只,二次随机分组:正常组10只、模型组16只、西药组14只、预防组14只、高量组14只、低量组14只。实验结束后,检测空腹血糖(FBG)、糖化血红蛋白(HbA1c)、尿微量白蛋白(mAlb)、β2微球蛋白(β2-MG)、肌酐(Scr)、尿素氮(BUN);HE及PAS染色观察病理学形态改变;免疫组化法观察肾组织中Bcl-2、Bax蛋白表达;实时荧光定量Q-PCR法检测肾组织中CTGF及ColⅣmRNA表达。结果:模型组FBG、HbA1c、mAlb、β2-MG、BUN明显升高(P<0.01),Scr无明显变化,肾脏病理改变明显,Bcl-2表达下降、Bax表达增加,CTGF、ColⅣmRNA含量明显上升(P<0.01),各治疗组上述指标明显改善。结论:益气解毒活络方可能通过抑制Bax表达,提高Bcl-2表达,降低mAlb,减少肾组织CTGF及ColⅣmRNA表达,从而保护肾脏。Objective： To study the effect of Yiqi Jiedu Huoluo Formula on the expression of connective tissue growth factor （CTGF） and Ⅳ collagen （ColⅣ） mRNA of rats with diabetic nephropathy （DN） in renal tissue induced by STZ. Methods： 90 Wistar rats were randomly twice divided into the follow groups： control group （n=10）, model group （n=16）, benazepril group （n=14）, prevention group （n=14）, high dose group （n=14）, low dose group （n=14）. After the experiment, the fasting blood glucose （FBG）, glycosylated hemoglobin （HbAlc）, urine microalbuminuria （mAlb）, 13 2-microglobulin （ 13 2-MG）, creatinine （Scr） and blood urea nitrogen （BUN） were tested. HE and PAS staining were used to evaluate the renal pathological changes. Immunohistochemical was used to observe the protein expressions of Bcl-2 and Bax. Real-time fluorescent quantitative PCR was used to test the expressions of CTGF and ColⅣ mRNA in kidney tissues. Results： The levels of FBG, HbAlc, mAlb, β 2-MG and BUN of rats in the model group increased obviously （P〈0.01）, and the level of Scr has no significant change. The pathologic change of renal tissue was obvious. The expression of Bcl-2 decreased, and expression of Bax increased. The expressions of CTGF and ColⅣ mRNA increased significantly （P〈0.01）. The above indexes of rats in treatment group improved obviously. Conclusions： Yiqi Jiedu Huoluo Formula can protect the kidney by regulating the expression of Bax, Bcl-2, mAlb and mRNA of CTGF and CollV.
