摘  要:目的:观察基质金属蛋白酶(MMP)-1、MMP-3、MMP-13和TIMP-1在实验性膝骨关节炎兔软骨中的变化规律。方法:改良后的Videman法左后肢伸直位固定制动法建立模型,通过针刀、电针和圆利针干预,造模后13周取材,采用RT-PCR和Western blot检测软骨MMP相关因子基因和蛋白表达水平。结果:造模后13周,MMP-1 mRNA各组无统计学差异。MMP-1蛋白表达:模型组较正常组显著升高(P<0.05);MMP-3,MMP-13 mRNA和蛋白表达,模型组与电针组较正常组明显升高(P<0.05);针刀组和圆利针组较模型组明显降低(P<0.05);针刀组和圆利针组较电针组显著降低(P<0.05);其中,MMP-3 mRNA圆利针组表达明显高于正常组(P<0.05),电针组明显低于模型组(P<0.05),TIMP-1 mRNA各组高于正常组(P<0.05),电针组显著高于模型组(P<0.05);TIMP-1蛋白表达模型组较正常组明显降低(P<0.05),干预各组较模型组显著升高(P<0.05)。结论:针刀和圆利针对MMP-3、MMP-13 mRNA和蛋白表达有抑制作用,表明其治疗机制可能与调节基质金属蛋白酶家族有关,这与电针治疗关节损伤的机制存在差异。Objective: To observe the variation regularity of the expression of MMP-1, MMP-3, MMP-13 and TIMP-1 of cartilage in rabbits with KOA. Methods: The left knee joint was fixed with piaster bandage in extention position to establish the rabbit model of knee joint injury with the modified videman method. Acupotomy (AP), electro-acupuncture (EA) and round- sharp acupuncture (RSA) were applied to treat in AP group EA group and RSA group. Expression of MMP-1, MMP-3, MMP-13, TIMP-1 mRNA by RT-PCR and expression of proteins by Western blot tests. Results: Expression of MMP-1 mRNA, there was no significant difference between groups; Expression of MMP-1 protein, in comparison with normal control group, model group increased significantly (P〈0.05). Expression of MMP-3, MMP-13 mRNA and proteins, in comparison with control group, model group, EA group increased significantly (P〈0.05); to compare with model group, AP group and RSA group decreased significantly (P〈0.05), to compare with EA group, AP group and RSA group decreased significantly (P〈0.05), and RSA group of MMP-3 mRNA is much higer than normal control group (P〈0.05), MMP-3 mRNA in EA group is much higer than model group (P〈0.05). In comparison with normal control group after modeling, TIMP-1 mRNA of model group, EA group, AP group and RSA group increased significantly (P〈0.05), and EA group is much higher than model group (P〈0.05). Expression of TIMP-1 protein, in comparison with normal control group, model group decreased significantly (P〈0.05), and AP group, EA group and RSA group are much higer than model group (P〈0.05). Conclusion: The Acupotomy and round-sharp acupuncture adjusting the MMps family may be one of the therapeutic mechanism,but the therapy mechanism are different from Electro-Acupuncture.
