半夏泻心汤对幽门螺杆菌诱导GES-1细胞凋亡及Bax表达的影响

摘　　要：目的:探讨半夏泻心汤对幽门螺杆菌(HP)感染胃黏膜上皮细胞GES-1凋亡的影响,以及对促凋亡基因Bax表达的作用。方法:实验分对照组、感染组和半夏泻心汤组,以1∶50的细胞:细菌比例感染GES-1细胞,共培养48h。对照组和感染组采用正常鼠血清培养,半夏泻心汤组用含药鼠血清培养。流式细胞仪检测细胞凋亡率。实时定量PCR检测Bax mRNA,Western blot检测Bax蛋白。结果:与对照组比较,感染组的细胞凋亡率显著升高(P<0.05),Bax mRNA和蛋白表达增加(P<0.05)。与感染组比较,半夏泻心汤组细胞凋亡率降低(P<0.05)、Bax mRNA和蛋白表达均下调(P<0.05)。结论:半夏泻心汤通过下调Bax基因表达,抑制感染细胞的凋亡,减轻了Hp对细胞的损伤。Objective： To investigate the effects of Banxia Xiexin Decoction on apoptosis and expression of Bax of GES-1 cells infected by Helicobacter pyloN. Methods： 3 groups, including control group, model group and Banxia Xiexin Decoction group, were applied in the study. Cells were infected by Hp at 1：50 rate of cells to bacteria in model group and Banxia Xiexin Decoction group. After infection, cells in Banxia Xiexin Decoction group were cultivated in Banxia Xiexin Decoction- containing serum culture for 48h, while other two groups in normal rat serum culture. Apoptosis was evaluated by flow cytometry. Bax mRNA was measured by real-time quantitative PCR, and the protein by Western blot. Results： Compared with control group, apoptosis rate, expression of Bax mP, NA and protein increased significantly in model group （P〈0.05）. Compared with control group, the three parameters decreased in Banxia Xiexin Decoction group （P〈0.05）. Conclusion： Banxia Xiexin Decoction can reduce the damage of Hp to GES-1 cells by down regulating Bax expression and cell apoptosis.
