摘  要:探讨共刺激分子在Pristane诱导狼疮鼠体内的表达变化及意义。采用6~8周龄雌性BALB/c鼠单次腹腔注射0.5ml Pristane,对照组注射0.5ml PBS。通过尿蛋白试纸法检测尿蛋白含量;ELISA法检测外周血自身抗体(抗ds-DNA、南Sm/RNP)含量;HE染色评估病理学损伤;流式细胞术检测小鼠外周血、脾脏淋巴细胞表面共刺激分子CD28、CD40L/D40、CDl37/CDl37I。的表达;免疫组织化学法检测小鼠组织中共刺激分子CD28、CD40L、CDl37的表达。实验结果点示:与对照组小鼠相比,模型组小鼠出现多种SLE样临床症状和体征;模型组小鼠外周血和脾脏淋巴细胞表面共刺激分jCD28,一个月时表达增高,后逐渐下降至低于正常对照组(P〈0.05);CD40三个月时表达升高,六个月时表达降低至于正常对照组(P〈0.05),而CD40I。的表达变化无统计学意义(P〉0.05);CDl37/CDl37L三个月时表达增高并维持高表达状态至处死(P〈O.05);狼疮鼠肺组织中共刺激分子CD28、CD40L和CDl37呈现高表达状态,其他组织中变化并不明显。以上结果提示,Pristane诱导狼疮鼠体内共刺激分子表达发生异常变化,可能与其疾病的发生发展有密切关系,为临床治疗系统性红斑狼疮提供了研究基础。To study the expression and significance of costimulatory molecules in Pristane-induced lupus model mice, single intraperitoneal injection of 0.5 ml Pristane or PBS was applied to 6~8 week old female BALB/c mice. Urine protein was detected by urine dipstick test, serum autoantibodies (anti-ds-DNA, anti-Sm/RNP) were detected by ELISA, and pathological damages in tissues were assessed by HE staining. Costimulatory molecules in the peripheral blood and the spleen, such as CD28, CD40L/CD40 and CD137/CD137L, were detected by flow eytometry. The costimulatory molecules in tissues were also detected by immunohistochemistry. Compared with the control group, model group mice appeared a variety of SLE clinical signs and symptoms. In peripheral blood and the spleen of the lupus group, CD28 increased at the first month, then began to decrease at the third month and after six months it reduced to a level lower than that of the control group(P〈0.05) ;CD40 increased at the third month, and after six months it also reduced to a lower than that of the control group(P〈0.05), in the meanwhile, however, CD40L had no significant change(P〉0.05). Both CD137 and CD137L increased at the third month, and kept high expression levels till the mice died(P〈0.05). In the lung tissue of the lupus mice, the expression of eostimulatory molecules CD28, CD40L and CD137 were higher than those of the control group, but, no change was found in the other tissues examined . The above results suggested that the costimulatory molecules in pristaneinduced lupus mice express abnormally, and may be closely related to the development of SLE. This research provides a theoretical basis for clinical therapies of SLE.
