摘 要:目的:测定变应性鼻炎患者外周血中神经生长因子(NGF)、脑源性神经营养因子(BDNF)、神经营养蛋白-3(NT-3)mRNA的表达水平,观察其表达与Th1/Th2免疫失衡的关系。方法:采用成组对照试验,使用实时定量RT-PCR检测外周血中NGF、BDNF、NT-3mRNA的表达水平,2-ΔΔCt法计算实验组基因表达的变化倍数。ELISA检测Th2类细胞因子和Th1类细胞因子的含量。结果:相较于正常成人,变应性鼻炎患者外周血中NGF、BDNF、NT-3mRNA的表达量是正常人的2.44、4.46和1.78倍,NT-3mRNA的表达随着鼻炎严重程度加重呈递增趋势。变应性鼻炎患者外周血中Th2类细胞因子IL-4、IL-6、IL-10的浓度依次为2198±472、9407±703、3917±323μg/L,明显高于正常成人(P<0.05);而Th1类细胞因子INF-a的浓度为176±15μg/L,明显低于正常成人(P<0.05)。NGF、NT-3的扩增水平与IL-4、IL-6、IL-10浓度呈正相关。结论:相较于正常成人,变应性鼻炎患者外周血中NGF、BDNF、NT-3mRNA的表达量上调,NGF、NT-3可能通过Th1/Th2免疫失衡促使变应性鼻炎发病。ObjectiveTo detect the expression of NGF,BDNF,NT-3 mRNA in the peripheral blood of patientswith allergic rhinitis(AR). And to analyze the correlation between NGF,BDNF,NT-3 mRNA expression and theepidsode of rhinitis through Th-2 Hypothesis. Method:This study was a group controlled trial. The expression ofNGF, BDNF and NT-3 mRNA were tested by real-time quantitative RT-PCR and the concentrations of IL-4, IL-6, IL-10 and INF-a were tested by ELISA. Result.. The expression of NGF, BDNF and NT-3 mRNA in AR pa-tients were 2.44, 4. 46 and 1.78 times the amount of those in the healthy adults, respectively. The increased ex-pression of NT-3 correlated positively with the scores of visual analog scale of AR. The concentrations of IL-4,IL-6 and IL-10, which were 2198 ± 472 μg/mL, 9407± 703 μg/mL and 3917 ±323 μg/mL respectively, werehigher than those in the healthy adults. The concentration of INF-a was 2198 ± 472 μg/mL and less than thehealthy adults. The increased expressions of NGF,NT-3 were positively related to the increase of IL-4, IL-6 andIL-10. Conclusion:The expressions of NGF, BDNF and NT-3 mRNA in AR patients are higher than those in thehealthy adults. NGF, BDNF and NT-3 may contribute to the pathogenesis of AR. Moreover, NGF and NT-3 mayinduce the episode of rhinitis through Th-2 Hypothesis.
