摘 要:目的:比较甘草泻心汤与其配方颗粒对复发性口腔溃疡(ROU)模型大鼠的药效学。方法:以免疫法复制大鼠ROU模型。40只Wistar大鼠随机均分为正常对照(等容生理盐水)组、模型(等容生理盐水)组、左旋咪唑(17.5 mg/kg)组、甘草泻心汤(12g/kg)组、甘草泻心汤配方颗粒(12 g/kg)组。灌胃给药,每天1次,连续20 d。酶联免疫吸附法检测大鼠血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-8的含量;流式细胞仪测定大鼠外周血T淋巴细胞亚群;热板法检测大鼠痛阈值。结果:与正常对照组比较,模型组大鼠血清TNF-α、IL-8含量增加,CD4+T细胞减少,CD8+T细胞增加,CD4+/CD8+比值降低,痛阈值降低,差异有统计学意义(P<0.01)。与模型组比较,甘草泻心汤组、甘草泻心汤配方颗粒组大鼠血清TNF-α、IL-8含量减少,CD4+T细胞增加,CD8+T细胞减少,CD4+/CD8+比值升高,痛阈值升高,差异有统计学意义(P<0.01),且甘草泻心汤组与甘草泻心汤配方颗粒组比较差异无统计学意义(P>0.05)。结论:甘草泻心汤及其配方颗粒具有抗炎、免疫调节和镇痛作用,对ROU模型大鼠均有较好的保护作用,两组间药效学相当。OBJECTIVE: To compare the pharmacodynamics between Glycyrrhizae decoction for purging stomach-fire and its formula granules on model rats with recurrent oral ulcer (ROU). METHODS: ROU model was induced by immunifaction. 40 Wistar rats were randomly divided into normal control group (constant volume of normal saline), model group (constant volume of normal saline), levamisole group (17.5 mg/kg), Glycyrrhizae decoction for purging stomach-fire group (12 g/kg) and formula granules of Glycyrrhizae decoction for purging stomach-fire group ( 12 g/kg). They were given medicine intragastrically once a day for consecutive 20 days. The contents of TNF-α and IL-8 were determined by ELISA. T lymphocyte subsets of peripheral blood was determined by flow cytometry and the pain threshold in rats was tested by hot plate method. RESULTS : Compared with normal con- trol group, the serum contents of TNF-α , IL-8 and CD8+ cells were increased in model group, while the number of CD4+cells and CD4+/CD8+ ratio and pain threshold were all decreased; there was statistical significance (P〈0.01). Compared with model group, the serum contents of TNF-α , IL-8 and CD8+ cells were decreased in Glycyrrhizae decoction for purging stomach-fire and its formula granules groups, while the number of CD4+ cells and CD47CD8+ ratio and pain threshold were all increased; there was statistical significance(P〈0.01); there was no statistical significance between Glycyrrhizae decoction for purging stomach-fire group and its formula granules group (P〉0.05). CONCLUSIONS: Both Glycyrrhizae decoction for purging stomach-fire and its formula granules have the effects of anti-inflammatory, immuno-regulation and analgesia, and both have good therapeutic effect on ROU model rats; no difference is found in pharmacodynamics between 2 groups.
