摘  要:目的:探讨金雀异黄素(GEN)对子宫内膜异位症(EMs)大鼠模型中雌激素受体(ER)α、β表达水平的影响。方法:将大鼠分为正常组(A组)和子宫内膜异位症组;子宫内膜异位症组随机分为4组:模型组(B组)、GEN低剂量组(C组)、GEN中剂量组(D组)和GEN高剂量组(E组)。测量血清中雌二醇(E2)的含量,通过RT-PCR方法检测种植物中ERα、ERβ在mRNA水平上的表达,免疫组化法观察异位内膜组织中ERα、ERβ的表达。结果:与治疗前比较,血清中E2的含量差异无统计学意义(P>0.05);高剂量GEN组子宫内膜异位模型中种植物组织中ERα、ERβmRNA水平下降;种植物中ERα的表达在各组之间无明显差异,但GEN中剂量和GEN高剂量治疗组中种植物的ERβ蛋白表达较模型组明显下降(P<0.01)。结论:GEN对EMs的抑制作用可能更趋向于ERβ选择性拮抗剂的作用。Objective: To explore the eitect of genistein on expression levels oi estrogen receptor (ER) α and ERβ m rat model ot endometriosis. Methods: The rats were divided into normal group (group A) and endometriosis group; the rats in endometriosis group were randomly divided into model group (group B), lose -dose genistein group (group C), middle -dose genistein group (group D) and high - dose genistein group ( group E) . The contents of serum estradiol in the five groups were detected, RT - PCR was used to detect the expression levels of ERα mRNA and ERβ mRNA, immunohistoehemieal method was used to observe the expression levels of ERα and ERβ in endometriosis. Results: There was no statistically significant difference in serum estradiol content between before treatment and after treatment ( P 〉 0.05 ) ; ERα mRNA and ERβ mRNA levels in high - dose genistein group decreased after treatment; there was no statistically signifi- cant difference in ERα expression level among the five groups, but ERβ expression levels in middle - dose genistein group and high - dose genistein group decreased significantly compared with model group (P 〈 0. 01 ) . Conclusion: The inhibiting effect of genistein on endometriosis tend to the effect of ERβ - selective antagonist.
