摘  要:目的:通过分析各组大鼠血液代谢物图谱,发现小青龙汤证血液的生物标志物,为临床诊断和治疗提供客观依据。方法:对空白对照组、模型组和小青龙汤组大鼠的尿液进行核磁共振氢谱检测,图谱经MestRec预处理和积分后,所得数据用SIMCA-P+进行偏最小二乘法判别分析分析(PLS-DA),得到相应的散点图和载荷图,进行比较后找出差异性代谢物。结果:空白对照组与模型组比较,模型组中肌酸、3-羟基丁酸、牛磺酸、天冬氨酸、瓜氨酸、谷氨酰胺升高(P<0.05),乳酸、醋酸(δ1.9)、酪氨酸、脯氨酸、甘氨酸降低(P<0.05),小青龙汤组经治疗后肌酸、牛磺酸仍升高,但和模型组相比有所降低(P<0.05),酪氨酸、脯氨酸仍低于空白对照组,但较模型组升高,瓜氨酸和谷氨酰胺在较模型组更高(P<0.05)。结论:血液中的代谢标志物主要与能量代谢紊乱、机体免疫力以及机体应激有关,尤其与能量代谢和免疫相关的代谢物变化较明显。Objective: Through the map analysis of various metabolites in rats blood,the biomarkers in syndrome suitable for Small Qinglong Decoction were found out,which provides objective basis for the clinical diagnosis and treatment.Methods: Based on the rats blood NMR spectrum detection of the blank control group,the model group and the Small Qinglong Decoction group's data came from the map by Mest Rec pretreatment and integration was analyzed using SIMCA-P + by( PLS-DA),and then,the corresponding scatter diagram and diagram of loading were got and the differences of metabolites were found out by the comparison. Result: The results from comparison of model group and the blank control group showed that the contents of metabolites of creatine,3-hydroxybutyric acid,taurine,aspartic acid,citrulline and glutamine were increased( P 0. 05). The contents of metabolites of lactic acid,acetic acid,proline,tyrosine and glycine were decreased( P 0. 05). In small Qinglong Decoction group,taurine was still higher than that in the blank control group but lower than that in the model group( P 0. 05). Tyrosine and proline were still lower than that of the blank control group,but were increased compared with that in the model group( P 0. 05),and the contents of metabolites of citrulline and glutamine were much higher than that in the model group. Conclusion: The micro marker materials were mainly related to energy metabolism,immune and the body's stress response,especially,relating energy metabolism and immune change obviously.
