半夏内生菌的分离及抗菌活性筛选

摘　　要：目的:分离三叶半夏和掌叶半夏的内生菌,并对分离的内生菌进行抗菌活性筛选。方法:采用组织块分离法从不同品种半夏中分离内生菌,并对分离所得内生菌进行初步鉴定。利用对峙法进行抗菌活性筛选,获得具有抑菌活性内生菌。结果:共分离得到内生菌39株,其中内生细菌15株、内生真菌24株。对峙法筛选实验结果表明:5株菌对大肠杆菌具有较强拮抗性,5株菌对金色葡萄球菌有较强拮抗性,4株菌对枯草杆菌有较强拮抗性。结论:半夏含有丰富的内生菌菌种资源,实验表明多株菌具有代谢抑菌活性物质的特性,在获取天然药用生物活性成分方面存在很大潜能。Objective ： The aim of this study was to isolate the endophyte of Pinellia pedatisecta Sehott and Pinellia ternate. To screen and identify Escherichia coli, staphylococcus aureus, bacillus subtilis antagonistic bacteria. Method ： Two types of Pinellia roots, stems and leaves were take out. After sterilization surface in MS medium vaccinations,the endogenous growth of bacteria was made. And then, the streaked plate was carried on to separate and purify endophytes. And the method of confrontation of endogenous was used to screen bacteria. According to the characteristics of train and separation of microorganism morphological characteristics, gram stain, etc, the endogenous antagonism bacteria were screened out and identified. Result ： Fifteen endophytic bacteria and twenty - four endophytic fungi were separated from the endophyte of two plants. The screened five endophytie bacteria with strong antagonistic effect on the Escheriehia eoli, five endophytic bacte- ria with strong antagonistic effect on the staphylococcus aureus ,four endophytic bacteria with strong antagonistic effect on the bacillus subtilis. Conclusion：Pinellia ternata contains rich resources of endogenous fungus strains. Experiments show that many strains of bacteria have metabolic antibacterial activity material characteristics. There is great potential about natural medicinal bioactive components.
