摘 要:目的:探讨四磨汤对寒冷束缚应激小鼠脑肠肽NOS的影响。方法:动物随机分组,采用寒冷束缚等多种刺激方法,制备胃肠功能紊乱小鼠模型,分别给予蒸馏水、得舒特和四磨汤口服液治疗;免疫组织化学法观察小鼠脑内NOS的阳性表达,用RT-PCR法检测结肠组织NOS mRNA表达。结果:模型组小鼠NOS阳性表达减少,平均OD值与正常组比较差异有显著性统计学意义(P<0.01);四磨汤组NOS表达增加,与模型组比较有统计学差异(P<0.05)。四磨汤可调控模型小鼠结肠组织Nos mRNA,与模型组比较差异有显著性统计学意义(P<0.01)。结论:四磨汤调节脑肠肽NOS的表达可能是其治疗功能性胃肠病的作用机制之一。Objective:To study the effects of Simo Decoction on expression of NOS in cold - constraint stressed mice. Methods:Gastrointestinal dysfunction model was established by exerting the factors of cold - constraint stimulation. And then they were randomly treated with distilled water, Dieetel and SD. The expressions of NOS in brain tissue were meas- ured by immunohistoehemieal staining. The expressions of NOS in intestine were measured by RT - PCR. Result : The ex- pressions of NOS in brain tissue of model group was decreased significantly compared with that of the normal group ( P 〈 0. 01 ). The SD could increase the expression of NOS in brain tissue. The average optical density of immune positive result had significant differences compared with that of the model group ( P 〈 0.05 ). The SD could regulate the expression of NOS mRNA ,which had significant differences compared with that of the model group (P〈 0.01 ). Conclusion:Simo Decoction appears potential to regulate the expression of NOS in cold - constraint stressed mice, which is one of possible mechanism to treat FGIDs.
