摘 要:目的:观察参麦汤对糖耐量低减大鼠白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及肝组织病理变化的影响。方法:高脂高糖饲料喂养结合腹腔注射小剂量链脲佐菌素溶液的方法复制糖耐量低减大鼠模型;用参麦汤治疗;实验结束后采用放射免疫法测血清TNF-α、采用酶联免疫(ELISA)法测定IL-6、处死动物后取肝组织常规石蜡包埋切片,HE染色,光镜下观察肝组织形态的变化。结果:和正常组比较,模型组TNF-α、IL-6的浓度升高,有显著性差异(TNF-αP<0.05,IL-6 P<0.05);与模型组比较,各干预组均可降低TNF-α、IL-6的浓度,其中以参麦汤组为著(P<0.05),参麦汤加运动组(P<0.05)、运动组(P<0.05)亦有明显差异。组织形态观察显示:糖耐量异常大鼠的肝组织存在脂肪样变;各治疗组对糖耐量异常大鼠肝组织的作用,以参麦汤加运动组改善较理想。结论:糖耐量低减期存在肝的脂肪样变;参麦汤可改善糖耐量异常大鼠的IL-6、TNF-α水平,并在一定程度上逆转病变肝组织。Objective:To observe the influence of Shenmai Decoction on impaired glucose tolerance (IGT) rats interleukin - 6 ( IL - 6) and tumor necrosis factor alpha ( TNF - α) and liver tissue pathological morphology. Methods : High fat and sugar diet combined with intraperitoneal injection of low dose of streptozotocin was used for replicating the impaired glucose tolerance rats' model. The treatment was Shenmai Decoction. After the experiment, radioimmunity method was used to measure the TNF - cx and enzyme - linked immune method was used to determine IL - 6. After death, liver tissues was taken out for conventional paraffin embedding slices. By HE staining, the liver tissue morphological change was observed. Results : Compared with normal group, model group' s TNF - α and IL - 6 levels increased and there was significant difference ( TNF - α,P 〈 0.05, IL - 6, P 〈 0.05 ). Compared with model group, the intervention group can reduce TNF - α and IL - 6 levels, of which the most significant role was in the Shenmai Decoction group ( P 〈 0.05 ), and Shenmai Decoction and exercise group also had obvious difference ( P 〈 0.05 ). Tissue morphology observation showed that abnormal glucose tolerance in rats of liver tissue had fatty degeneration. Shenmai Decoction and exercise group can improve the impaired glucose tolerance ideally. Conclusion :There is liver tissue fatty degeneration of IGT rat. And Shenmai Decoction can improve IL- 6 and TNF -α levels of TGT rats, and to a certain extent, reverse the pathological changes of liver tissue.
