益气活血方对胃癌前病变大鼠NF-κB、cyclinD1和p16表达的影响

摘　　要：目的:观察益气活血方对胃癌前病变大鼠NF-κB、cyclinD1和p16表达的影响。方法:64只SD大鼠随机分为正常对照组8只和造模组56只,采用以MNNG损伤为主的综合法复制大鼠胃癌前病变模型,随机处死10只大鼠经病理确定造模成功后,再将46只造模组大鼠随机分为模型组12只、益气活血高、低剂量组各12只和胃复春组10只,分别给予益气活血方、胃复春混悬液灌胃治疗12周。免疫组化Envision法测定大鼠NF-κB、cyclinD1和p16的表达。结果:与正常对照组比较,模型组大鼠胃组织NF-κBp65/IκBα表达明显升高(P<0.01)。药物治疗后,益气活血高剂量组、胃复春组大鼠胃组织NF-κBp65表达较模型组明显降低(P<0.05),IκBα蛋白表达较模型组明显升高(P<0.01)。与正常对照组比较,模型组大鼠胃组织cyclinD1和p16明显升高(P<0.01,P<0.05);药物治疗后,益气活血高剂量组及胃复春组大鼠胃组织cyclinD1表达较模型组比较明显降低(P<0.01,P<0.05);益气活血高剂量组大鼠胃组织p16表达较模型组明显升高(P<0.01)。结论:益气活血法可能通过减少致炎因子激活的NF-κB信号通路抑制cyclinDl,上调p16表达防治胃癌前病变,这可能是其防治胃癌前病变的作用机制之一。Objective：To investigate the impact of Yiqi Huoxue therapies on the expressions of nuclear factor - κB （ NF - κB）, cyclinD1 and p16 in rats with gastric precancerous lesions. Methods ： Sixty - four SD rats were randomly divided into blank control group （ n = 8 ） and model control group （ n = 56 ）. Gastric precancerous lesions were induced in rats by MNNG injury to the stomach. Ten model rats were randomly sacrificed for histopathological examination. The rest 46 model rats were divided into four groups：model group （ n = 12） ,high dose of Yiqi Huoxue group （ n = 12） ,low dose of Yiqi Huoxue group （n = 12 ） and Weifuchun group （n = 10）. The corresponding drugs were intragastrically given for 12 weeks. The expressions of NF - κB, cyclinD1 and p16 were observed by Envision. Results：Compared with the blank control group, the expressions of NF - KBp65/IκBα were significantly greater （P 〈 0.01 ） in the model group. After Yiqi Huoxue and Weifuchun treatment, the expression of NF - κBp65 was decreased in high lose of Yiqi Huoxue group and Weifuchun group （P 〈 0.05 ） and the expression of IκBα was increased （P 〈 0.01 ） as compared with that of the model control group. Compared with the blank control group, the expressions of cyclinD1 and p16 were significantly greater （P 〈 0.01, P 〈 0.05 ） in the model group. After Yiqi Huoxue and Weifuchun treatment, the expression of cyclinD1 was reduced signif- icantly in high dose of Yiqi Huoxue group and Weifuchun group（P 〈 0.01, P 〈 0.05 ）and the expression of p16 in high dose of Yiqi Huoxue group was increased （P 〈 0.01 ） as compared with those of the model control group. Conclusion ： Yiqi Huoxue therapies can significantly adjust the expressions of NF - κB to inhibit the level of cyclinD1 and improve p16 in rats ,which may be one of the mechanisms underlying the therapeutic effect of Yiqi Huoxue against gastric precancerous lesions.
