摘 要:目的痘苗病毒作为疫苗载体具有接种途径简单、高滴度、低成本等优点,重组痘苗病毒投入使用之前进行理化特性试验,对重组痘苗病毒的生产、运输和保存有一定重要意义。方法将重组痘苗病毒rVTT-JEVE作热、酸、碱、乙醚、氯仿、胰蛋白酶、超声波以及紫外线处理,然后接种到BHK-21细胞上,观察rVTT-JEVE毒价的变化,分析理化因素对rVTT-JEVE的影响。结果在pH3~9范围内,酸碱度变化不会引起rVTT-JEVE毒价的显著变化。rVTT-JEVE经50℃处理60min、30min和60℃处理15min可被灭活;对5%的氯仿敏感;经终浓度0.5%的胰蛋白酶37℃作用1h,病毒毒价降低2个滴度;对25%的乙醚不敏感;经40KHz超声波处理60min和80KHz处理30min即可灭活;30W紫外灯垂直距离10cm照射15min、30cm照射30min即可灭活。结论 rVTT-JEVE耐热、酸、碱、乙醚、氯仿及胰蛋白酶,对超声波、紫外线较敏感。Objective As a vaccine vector,the vaccinia virus has many advantages,including a simple route of inoculation,a high titer,and a low cost.Before a recombinant vaccinia viruse is used as a vector,it must undergo physical and chemical tests that are key to the production,transportation,and preservation of recombinant vaccinia viruses. Methods After treatment with heat,acids,alkalis,ether,chloroform,trypsin,ultrasonic waves,and ultraviolet rays,the recombinant vaccinia virus rVTT-JEVE was inoculated into BHK-21 cells and then changes in the viral titer of rVTT-JEVE were observed.The impact of these physical and chemical factors on rVTT-JEVE was then analyzed. Results Results indicated that changes in pH did not cause significant changes in the viral titer of rVTT-JEVE in a pH range of 3.0-9.0.Subjection to a temperature of 50 ℃for 60 min and 30 min and a temperature of 60 ℃for 15 min inactivated rVTTJEVE.rVTT-JEVE was sensitive to 5%chloroform.Subjection to 0.5%trypsin at 37℃for 1hreduced the viral titer by 2logs.rVTT-JEVE was not sensitive to 25%ether.Subjection to ultrasonic waves at 40 kHz for 60 min and at 80 kHz for 30 min inactivated rVTT-JEVE.Subjection to ultraviolet rays at a vertical distance of 10 cm for 15 min and at 30 cm for 30 min inactivated rVTT-JEVE. Conclusion The characterization of rVTT-JEVE to heat,acid,alkali,ether,chloroform,trypsin and the sensitivity to ultrasonic,ultraviolet were identified.
