摘  要:目的了解wnt信号通路与宫颈癌发生的关系,为该病的早期诊断及治疗提供指导,并为其靶向治疗提供新思路。方法采用免疫组织化学法分别检测正常宫颈组织、宫颈上皮内瘤变组织、宫颈鳞癌组织中wnt-3a和β-catenin的表达状况,分析其与疾病的相关性;采用RT-PCR法检测上述3种组织中wnt-1和wnt-3a的mRNA表达水平。结果wnt-3a蛋白在正常宫颈组织、宫颈上皮内瘤样变和宫颈鳞癌组织表达率分别为10.0%、37.5%和39.3%,表达发生在细胞浆细胞中,β-catenin蛋白在正常宫颈鳞状上皮组织细胞膜上呈连续表达状态,主要分布在细胞膜的棘层和上皮基底层,而在宫颈上皮内瘤变及宫颈鳞癌组织细胞膜呈现不同程度的表达缺失,甚至不表达,其异常表达率分别为15.0%、50.0%和82.1%;wnt-3a蛋白的表达与β-catenin蛋白的异常表达呈正相关(r=0.195,P<0.05)。RT-PCR产物电泳显示wnt-1基因大小约100bp,wnt-3a基因约147bp。结论宫颈癌的发生发展与wnt信号通路的激活、β-catenin的表达异常密切相关,对wnt-3a和β-catenin进行联合检测可用于宫颈癌的早期诊断。Objective To ascertain the relationship between the incidence of cervical cancer and the wnt signaling pathway in order to provide guidance for the early diagnosis and treatment of this disease and to provide new ideas for targeted therapy. Methods The expression of wnt-3aandβ-catenin was immunohistochemically detected in normal cervical tissue,cervical intraepithelial neoplasias,and cervical squamous cell carcinomas.The association between that expression and disease was analyzed and then levels of expression of wnt-1and wnt-3amRNA in the 3types of tissues were determined. Results wnt-3aprotein was slightly expressed(10%)in normal cervical tissue,where it appeared in the cell cytoplasm.The protein stained yellow in the cytoplasm of cervical intraepithelial neoplasias and cervical squamous cell carcinomas,where the corresponding rates of expression were 37.5%and 39.3%.In normal cervical squamous epithelial tissue,β-catenin protein was constantly expressed on the cell membrane,and this protein was mainly located in the membrane of cells in the stratum spinosum and basal layer of the epithelium.The protein was stained brown and the cytoplasm was clear.Cervical intraepithelial neoplasias and cervical squamous cell carcinomas exhibited a loss of expression at a rate of 15%,a lack of expression at a rate of 50%,and abnormal expression at a rate of 82.1%,those these rates varied.Expression of wnt-3aprotein and abnormal expression ofβ-catenin protein were correlated(P〈0.05,r=0.195).Electrophoresis revealed a target band at around 100 bp for the wnt-1gene and a target band at around 147 bp for the wnt-3a gene. Conclusion The development of cervical cancer is closely related to activation of the wnt signaling pathway and abnormal expression ofβ-catenin.Detection of both wnt-3aandβ-catenin expression can more accurately diagnose cervical cancer at an early stage.
