摘  要:目的:研究不同实验条件下,新物体识别实验(NOR实验)在ICR小鼠学习记忆能力评价中的应用效果.方法:将130只ICR雄性小鼠随机分为10组:4组用于不同间隔时间(10 min、90 min、4h、24 h)的测试,4组用于不同物体材质(木质-木质、塑胶-塑胶、塑胶-木质、木质-塑胶)的测试,2组用于重复测试(隔1d或3d测1次,共测3次).视频跟踪记录小鼠在开场内的活动轨迹,分析其探索新旧物体的时间,计算小鼠对新物体的分辨比(DR)和分辨指数(DI),评价各组小鼠的学习记忆情况.结果:检测间隔时间为10 min和90 min组的小鼠对新物体的DR和DI较旧物体显著加大(均P<0.01);新旧物体不同材质对小鼠探究物体的时间、DR和DI影响较大,选择同一材质物体可使小鼠对新物体保持较为稳定的DR和DI;NOR实验可在同一批小鼠中重复测试,小鼠可持续保持对新物体的探究兴趣.结论:进行NOR实验时,选择较短的检测间隔时间和选用相同材质的物体可较好地评价小鼠学习记忆能力.NOR实验可重复测试.Objective:To investigate the efficacy of novel object recognition (NOR) test in assessment of learning and memory ability in ICR mice in different experimental conditions.Methods:One hundred and thirty male ICR mice were randomly divided into 10 groups:4 groups for different inter-trial intervals (ITI:10 min,90 min,4 h,24 h),4 groups for different object materials (wood-wood,plastic-plastic,plasticwood,wood-plastic) and 2 groups for repeated test (measured once a day or every 3 days,totally three times in each group).The locomotor tracks in the open field were recorded.The amount of time spent exploring the novel and familiar objects,the discrimination ratio (DR) and the discrimination index (DI) were analyzed.Results:Compared with familiar object,DR and DI of novel object were both increased at ITI of 10 min and 90 rain (P 〈0.01).Exploring time,DR and DI were greatly influenced by different object materials.DR and DI remained stable by using identical object material.NOR test could be done repeatedly in the same batch of mice.Conclusion:NOR test can be used to assess the learning and memory ability in mice at shorter ITI and with identical material.It can be done repeatedly.
