摘 要:目的根据中医组方配伍雷公藤多苷(GTW)组成雷至胶囊及桑椹液,旨在对抗GTW的生殖毒性。方法4周龄Wistar大鼠分7组,正常对照组(自由进食水),雷公藤多苷高剂量组(20mg·kg^-1·d^-1),雷公藤多苷低剂量组(10mg·kg^-1·d^-1),雷至灌胃液高剂量组(RZ液,雷公藤多苷20mg·kgll·d.1+稀释后RZ液2ml/只),雷至灌胃液低剂量组(RZ液,雷公藤多苷10nag·kg^-1·d^-1+稀释后RZ液2rnl/只),桑椹液高剂量(雷公藤多苷20nag·kg^-1·d^-1+稀释后的桑椹液2ml/只),桑椹液低剂量(雷公藤多苷10mg·kg^-1·d^-1+稀释后的桑椹液2ml/只)。在给药前、给药30d各测定1次血常规、尿常规、血生化、血清性激素。给药30d与停药15d处死大鼠,取材组织病理检查。结果给药前、后各组血常规、肝肾功能、尿蛋白无明显变化。GTW给药1d大鼠睾酮下降,雷至胶囊组睾酮恢复明显,桑椹组改善不明显。组织病理可见,给药1个月雷公藤多苷、雷至灌胃液及桑椹液在所设剂量下均可见肾脏肾小管上皮水样变性、睾丸曲精小管各级生精上皮细胞减少、附睾管中精子明显减少并可见多核巨细胞,间质纤维结缔组织增生。停药半个月后,雷公藤多苷在所设剂量下可见肾脏肾小管上皮水样变性,睾丸曲精小管各级生精上皮细胞减少,附睾管中精子明显减少并可见多核巨细胞,间质纤维结缔组织增生。雷至灌胃液在所设剂量下可见附睾管中精子明显减少并可见多核巨细胞。桑椹液在所设剂量下可见睾丸曲精小管各级生精上皮细胞减少,附睾管中精子明显减少并可见多核巨细胞。结论单独给予雷至灌胃液对正常大鼠生殖系统可能影响不显著,仅在GTW造成生殖系统损害时发挥对抗作用,故可认为雷至胶囊的对抗作用机制为保护睾丸生精上皮免受GTW的损害,并使血清睾酮水平维持于正Objective According to the principles of formulating prescription of traditional Chi-nese medicine, the RZ capsule and Mulberry liquid have been made composed of glycosides of triptery giumwilfordii Hook. f ( GTW ) , with the aim to reducing the reproductive toxicity of GTW. Methods 4 weeks-old male Wistar rats were divided into 7 groups: control group, GTW at highdose group (GTW 20 mg·kg^-1·d^-1) ,GTW at low-dose group(GTW 10 nag. kg^-1·d^-1) ,RZ liquid at high-dose group(GTW 20 mg·kg^-1·d^-1 with RZ liquid 2 ml/d). RZ liquid at low-dose group(GTW 10 mg with RZ liquid 2 ml/d). Mulberry(SS solution)at high-dose group(GTW 20 mg·kg^-1. d^-1 with mulberry liquid 2 ml/d)and Mulberry(SS solution) at low-dose group (GTW 10 mg. kg^-1 · d^-1 with mulberry liquid 2 ml/d). 24 h urine and blood sample had been collected for detecting proteinuria, blood cell anal-ysis,biochemistry and serum sex hormone pretreatment and in 30 d post-treatment respectively. Allrats were put to death for histopathologic examination in 30 d post-treatment and 15 d post-treatment respectively. Results There were no statistically significant changes in blood cell analysis, hepatorenal function and proteinuria among groups pre-and post-treatment. Serum testosterone declined in GTW group rats, while serum testosterone improved in RZ liquid group statistically significantly, but the ser-um testosterone still kept low in SS group rats. Histopathologic injuries had been seen in all GTW,RZ and SS groups, including hydropic degeneration in renal tubular epithelial, the numbers of epithelial cells at all levels of testicular seminiferous and sperm in the epididymis decreasing significantly, but the numbers multinucleated macrophages and interstitial connective tissue cells were in increment. Hydrop-ic degeneration were seen in renal tubular epithelium in GTW group 15 d after stopping treatment, and the numbers of epithelial cells in testicular seminiferous at all levels was falling.
