系统性红斑狼疮中白介素-27/骨桥蛋白调控轴在Th17细胞异常中的作用

摘　　要：目的通过研究系统性红斑狼疮(SLE)患者中白介素-27(IL-27)和骨桥蛋白(OPN)对Th17细胞分泌白介素-17(IL-17)的影响,探讨IL-27/OPN调控轴在SLE Th17细胞异常中的重要作用。方法取50例SLE患者和40例健康对照组血清,用酶联免疫吸附试验法(ELISA)检测各组血清中IL-27,OPN和IL-17的表达水平,并分析各细胞因子的组间差异以及它们间的相互关系。结果 1活动期SLE患者的IL-27,OPN和IL-17表达显著高于非活动期SLE患者和健康对照组,差异有统计学意义(P均<0.01);非活动期SLE患者和健康对照组间IL-27,OPN和IL-17水平的差异无统计学意义(P均>0.05);2血清中IL-27与IL-17水平、OPN与IL-17水平之间呈明显正相关(P均<0.01);3SLE患者的疾病活动程度评分与血清中OPN,IL-17呈明显正相关(P均<0.01)。SLE患者的疾病活动程度评分与血清中IL-27无明显相关(P>0.05)。结论 IL-27/OPN调控轴在SLE患者Th17细胞异常中起着重要的作用。Objective To investigate the regulatory role of interleukin-27/osteopontin regulatory axis in abnormal Thl7 cell and to study the influence of IL-27 and OPN on IL-17 level in serum from SLE patients. Meth- ods The level of IL-27, OPN and IL-17 in serum from 40 healthy controls and 50 SLE patients were de- tected by enzyme linked immunosorbent assay methods （ELISA）. Results .The levels of IL-27, OPN and IL-17 were dramativally increased in serum of active SLE patients than that of inactive SLE patients and healthy controls （P 〈 0.01 ） , while no differences between inactive SLE patients and healthy controls were observed （P 〉 0.05）. （2）Significant positive correlation was shown between IL-27 and IL-17, OPN and IL-17 level in serum from SLE patients （ P 〈 0.01 ）. （3）There was significant positive correlation be- tween SLEDAI and the level of OPN and IL-17 in SLE patients（ P 〈 0.01 ）, while no significant correla- tion between SLEDAI and IL-27 （P 〉 0.05 ）. Conclusion IL-27/OPN regulatory axis plays an important role in the abnormal functions of Thl7 cell in SLE patients.
