复荣通脉胶囊对糖尿病大鼠心肌微血管内皮的影响

摘　　要：目的观察经验方复荣通脉胶囊对糖尿病大鼠心肌微血管内皮的干预作用,为防治糖尿病心肌病变提供可靠的试验依据。方法采用腹腔注射链脲佐菌素(STZ)法建立糖尿病大鼠模型,按完全随机原则分为正常组(Normal)、糖尿病模型组(DM)、复荣通脉胶囊低(FRL)、中(FRM)、高剂量(FRH)治疗组。中药治疗组大鼠连续治疗8周后,尾静脉取血测血糖;双抗夹心酶联免疫吸附法(ELISA)定量测定可溶性血管内皮细胞蛋白C受体(sEPCR)和可溶性血栓调节蛋白(sTM)浓度。结果治疗前造模各组大鼠血糖与正常组相比,均显著升高(P<0.01),造模各组间血糖差异无统计学意义。经灌胃治疗8周后,造模各组血糖与治疗前相比无明显差异,造模各组间血糖相比无差异;造模各组与正常组相比,sEPCR及sTM水平均显著升高(P<0.01),复荣通脉胶囊低剂量治疗组与糖尿病模型组相比,sEPCR、sTM水平均无显著改变,复荣通脉胶囊中剂量治疗组与糖尿病模型组及复荣通脉胶囊低剂量治疗组相比,sEPCR及sTM水平均明显降低(P<0.01)。复荣通脉胶囊高剂量治疗组与糖尿病模型组及复荣通脉胶囊低剂量治疗组相比,sEPCR及sTM水平均明显降低(P<0.01),与复荣通脉胶囊中剂量治疗组相比,sEPCR及sTM水平均无显著改变。结论复荣通脉胶囊无调节血糖作用,但可减轻内皮细胞损伤,有助于减少糖尿病心肌病的发生与发展。Objective To observe the intervention effect of Furongtongmai Capsule on myocardial microvascular endothelia in diabetic rats to provide the reliable experimental evidence for preventing and treating diabetic cardiomyopathy. Methods The intraperitoneal injection of streptozotocin （STZ） was adopted to establish the diabetic rat model. The model rats were randomly divided into the diabetic model group（DM）, Furongtongmai Capsule low（FRL）, middle（FRM）, high（FRH） dose treatment group. After 8 - week treatment, the rats in the Chinese medicine treatment group rats were performed the caudal vein blood collection and the blood glucose was detected. Soluble en-dothelial protein C receptor （sEPCR） and soluble thrombomodulin （sTM） concentration were measured quantitatively with double anti-body sandwich enzyme linked immunosorbent assay （ELISA） . Results The blood glucose level before treatment in the model groups was significantly higher than that in the normal control group（ P ﹤ 0. 01 ）, but the blood glucose level had no difference among various model groups; compared with the normal group, the sEPCR and sTM levels in the model groups were significantly increased（ P ﹤ 0. 01）, compared with the model group, the sEPCR and sTM levels in the FRL group had no significant change; compared with the model group and the FRL group, the sEPCR and sTM levels in the FRM group were significantly decreased. Compared with the model group and the FRL group, the sEPCR and sTM levels in the FRH group were significantly decreased（ P ﹤ 0. 01 ）, compared with the FRM group, the sEPCR and sTM levels in the FRH group had no significant change. Conclusion Furongtongmai Capsule has no effect for regulat-ing blood glucose, but can relieve the vascular endothelial cell injury and conduces to reduce the occurrence and development of dia-betic myocardial disease.
