摘  要:目的观察覆盆子酮对HepG2细胞糖消耗的影响及胰岛素信号转导通路中SHP-1和人胰岛素受体底物(IRS-1)表达的影响。方法体外培养人肝癌HepG2细胞,采用MTT法检测细胞活力;葡萄糖氧化酶法检测细胞糖消耗;RT-PCR法检测蛋白酪氨酸磷酸酶SHP-1的基因表达;Western-blot法检测IRS-1的表达量。结果覆盆子酮可明显促进HepG2细胞对葡萄糖的消耗,明显降低HepG2细胞SHP-1 mRNA基因表达量,并能促进IRS-1蛋白表达量。结论覆盆子酮可能通过影响胰岛素信号转导通路中IRS-1和SHP-1的表达发挥降糖作用。AIM To explore the effect of raspberry ketone on the glucose consumption and the expression of SHP-1 and insulin receptor substrate-1 ( IRS-1 ) in insulin signaling pathway of HepG2 cells. METHODS MTT assay was used to measure the cell viability of the HepG2 cells cultured in vitro. The glucose oxidase test was used to measure the glucose consumption of these cells. RT-PCR method was adopted to detect mRNA expression of SHP-1 gene and Westen-blot method was adopted for the measurement of IRS-1. RESULTS Raspberry ketone could obviously promote glucose consumption in HepG2 cells and increase the IRS-1 protein expression while de- crease SHP-1 mRNA gene expression of HepG2 cells. CONCLUTION The hypoglycemic mechanism of raspber- ry ketone is associated with the expression of SHP-1 and IRS-1 in insulin receptor signaling pathway.
