摘 要:目的:以药典规定的吴茱萸碱、吴茱萸次碱和柠檬苦素含量为指标,研究吴茱萸中药材最佳提取工艺。方法:应用单因素试验结合响应面优化分析法(response surface methodology),对吴茱萸提取工艺进行优化。在单因素试验基础上选取因素不同水平,根据Box-Behnken Design试验设计原理,采用3因素3水平的响应面分析法确定最佳提取工艺。结果:以吴茱萸碱、吴茱萸次碱和柠檬苦素含量为指标,吴茱萸最佳提取工艺为醇体积分数76.82%,超声功率100Hz,提取时间90min。结论:采用响应面-多指标评价法优化的吴茱萸提取工艺,方法简便、预测性良好。优化的提取工艺结果可靠、评价指标可控,可以用于规范吴茱萸的提取工艺与质量控制。Objective: To establish an optimal extraction process for Evodiae Fructus with the content of evodiamine, rutaecarpine and limonin as indexes. Methods: The single factor experiment combined with response surface methodology was performed to optimize the process extracting Evodiae Fructus. Based on single factor experiments, different optimum conditions for the extraction of evodiamine, rutaecarpine and limonin from Evodiae Fructus were selected. The method of response surface analysis with 3 factors and 3 levels was adopted according to the Box-Benhnken experimental design principles. Results: The optimum conditions for extraction of evodiamine, rutaecarpine and limonin from Evodiae Fructus were as follows: Evodiae Fructus was extracted by 76.82% ethanol with ultrasonic power of 100 Hz for 90 min. Conclusion: The method has advantages of simple operation and good predictive, and can be used for quality control of Evodiae Fructus.
