摘  要:目的探讨p38抑制剂SB203580(SB)联合G-CSF(粒细胞集落刺激因子)对2Gy全身照射免疫细胞的影响。方法将30只C57雄性小鼠分为对照组、照射组和联合给药组。对照组接受假照射,照射组和联合给药组接受2Gy全身照射。联合给药组G-CSF 1μ只腹腔注射给药,当天照射后2 h和6 h给药,连续给药5 d,共给药10次。SB15 mg/kg腹腔注射给药,照射24 h第一次给药,隔日给药,共给药5次。照射后10 d处死小鼠取材,外周血白细胞进行CD4、CD8、B220检测,胸腺细胞进行CD4和CD8检测,骨髓细胞进行ROS(活性氧)水平检测。结果与对照组比较,单纯照射组白细胞计数和B220比例下降,而外周血CD4细胞比例、胸腺细胞CD4CD8细胞比例及骨髓细胞ROS水平显著增加,差异有统计学意义(P0.05)。联合给药组外周血淋巴细胞比例和胸腺细胞CD4CD8细胞比例升高,骨髓ROS水平下降,与照射组相比差异有统计学意义(P0.05)结论p38抑制剂联合G-CSF对辐射引起的免疫细胞损伤有一定保护作用,意义有待进一步探讨。Objective The study was conducted to explore the effect of p38 inhibitor SB203580 (SB) combining with granulocyte colony-stimulating factor (G-CSF) on 2Gy total body irradiated (TBI) immune cells. Methods Thirty C57 male mice were divided into a control group, an irradiation group, and a medication group. The control group was given sham radiation, and the radiation and medication groups were subjected to 2Gy TBI. The medication group was intraperitoneally injected with 1 μg of G-CSF per mouse 2 h and 6 h after irradiation for 5 consecutive days (10 injections in total), as well as 15 mg/kg of SB 24 h after irradiation once ever two days (5 injections in total). The mice were sacrificed 10 days after irradiation, and peripheral blood leucocytes of the mice were isolated for CD4, CD8 and B220 detection. Thymic cells and bone marrow cells were also isolated for CD4 and CD8 detection and ROS level determination, respectively. Results As compared with the control group, the irradiation group showed a decrease in white blood cell count and B220 ratio and a significant increase in CD4 cell ratio in peripheral blood, CD4+ and CD8+ cell ratios in thymic cells, and ROS level in bone marrow cells (P〈0.05). As compared with the irradiation group, the medication group showed an increase in lymphocytes in peripheral blood and CD4+ and CD8+ cell ratios in thymic cells, and a decrease in ROS level in bone marrow cells (P〈0.05). Conclusion p38 inhibitor together with G-CSF is shown to protect against radiation-induced damage of immune cells. Further studies on the significance of the approach are required.
