摘 要:采用荧光定量PCR方法和免疫组化方法,研究了Wnt5a基因在鹅胚胎期皮肤定量表达及定位表达的规律,并运用显微测微尺测量毛囊深度。结果表明:Wnt5a基因在胚胎期12~20 d均有表达,其中12~20 d表达量持续上升,20 d表达量最高,20 d以后表达量开始降低,但趋于稳定。Wnt5a基因在13 d的表皮、真皮及羽芽处表达,18 d在羽枝嵴和毛乳头大量表达,23 d在初级毛囊的毛乳头和毛囊干细胞处有少量表达,在次级毛囊中少量表达,29 d初级毛囊形态发生基本完成,Wnt5a基因几乎不表达,在次级毛囊中表达。毛囊深度结果表明:14~20 d,毛囊深度缓慢增加,但是深度增加不明显,Wnt5a抑制毛囊生长发育;20~26 d,是毛囊深度发育的高峰,Wnt5a含量降低,对初级毛囊发育没有抑制作用;26 d以后,深度几乎没有变化。次级毛囊18 d开始发育;18~20 d,次级毛囊深度发育缓慢,高表达量Wnt5a抑制次级毛囊发育;此后Wnt5a表达量降低,次级毛囊深度不断增加。Using quantitative PCR and immunohistochemistry method, the pattern of quantitative expression and locating expression of Wnt5a gene in goose skin of embryonic period was studied, and follicle depth was measured with Microscope Graticules. The results showed that WntSa gene in 12 to 30 d of the embryonic period was all expressed, among them, the expression continued to rise from 12 to 20 d, the expression of 20 d was the highest, after 20 d, the expression index began to drop, but it tended to be stable. Wnt5a gene appeared in 13 d epidermis, corium and feather germ; a large amount of expression appeared in 18 d barb ridge and hair papilla; Wnt5a gene not only expressed slightly in 23 d hair papilla and hair follicle stem cella of primary follicles but also in secondary follicles; on 29 d, morphogenesis of primary follicles basically completed, WntSa gene hardly expressed, but in secondary follicles it was expressed. Follicle depth showed that from 12 to 20 d the depth of follicles increased slowly, but the increase was not obvious, and the growth and develop- ment of follicles were inhibited by Wnt5a gene ; from 20 to 26 d it was the peak of follicle depth de-velopment, Wnt5a gene levels decreased, and it had no inhibition effect on primary follicles; after 26 d there was very little change in depth; secondary follicles began to develop in 18 d, from 18to 20 d, depth development of secondary follicles was slow, high expression quantity of Wnt5a inhibited the development of secondary follicles, after that the expression quantity of Wnt5a went down, and the depth of secondary follicles increased constantly.
