白纹伊蚊气味受体基因OR21α的克隆及定量分析

摘  要:本文克隆白蚊伊蚊气味受体基因OR21a分析其是否与搜寻宿主或吸血行为相关。通过设计简并引物,PCR扩增得到白纹伊蚊气味受体基因OR21a段,采用SYBR荧光定量PCR方法,以自纹伊蚊β-肌动蛋白基因作为内参,比较了OR21a因在未吸血雌蚊、雄蚊、打断吸血雌蚊和饱血雌蚊头部表达量的差异。结果显示,白纹伊蚊OR21a因在未吸血雌蚊头部表达量最高,饱血雌蚊头部表达量最低,而在雄蚊头部的表达量介于打断吸血雌蚊和饱血雌蚊之间。本研究发现白蚊伊蚊会依据不同的生理需求改变OR21a因的表达量,推测可能与白纹伊蚊搜寻宿主及吸血行为有一定的相关性。Sequence of the odorant receptor gene (OR21a ofAedes albopictus were identified by using degenerate primers. Expression of OR21amRNA was investigated by quantitative Real-Time RCR in the heads of adult male, interrupt sucking female, engorged and non-feeding adult female. Levels of OR21amRNA were normalized to mRNA encoding 15-action. The results showed that OR21a were differentially expressed in the heads of adult male, feeding-interrupted female, engorged and non-feeding adult female. The expression level of non-feeding adult female, feeding-interrupted female, adult male and engorged adult female was reduced successively. The expression level of OR21a changed for need. Maybe the OR21a gene has relationship with the behavior of blood feeding.
