摘  要:目的探讨TGF-β1对哮喘气道平滑肌细胞增殖的影响及caveolin-1在TGF-β1诱导的哮喘大鼠气道平滑肌细胞增殖中的抑制作用。方法离体培养大鼠气道平滑肌细胞并进行分组,用CCK-8法检测1、10、100μg/L的TGF-β1作用后及ERK通路特异性抑制剂PD98059干预后各组ASMCs的增殖情况,用Western blot法检测PD98059及β-环糊精干预后caveolin-1、p-ERK1/2蛋白表达的情况。结果 10μg/L的TGF-β1刺激ASMCs增殖的作用最为显著(P<0.05),PD98059干预后,ASMCs增殖明显减少(P<0.05)。TGF-β1刺激ASMCs增殖过程中,caveolin-1蛋白表达量减少,p-ERK1/2的表达增加(P<0.05);使用β-环糊精破坏微囊的结构后,caveolin-1表达量减少,而p-ERK1/2表达量增加(P<0.05)。结论 caveolin-1可以抑制TGF-β1诱导的ASMC增殖,这种抑制作用可能是通过抑制ERK1/2通路来实现的。Objective To investigate the effect of the transforming growth factor-β1 (TGF-β1) on airway smooth muscle ceils (ASMCs) proliferation and the repressive effect of caveolin-1 on TGF-β1-induced ASMCs proliferation of asthmatic rats.Methods Airway smooth muscle cells in vitro of rats were cultured.CCK-8 method was used to detect cell proliferation and Western blot method was used to detect the expression of caveolin-1 and p-ERK1/2 protein.Results CCK-8 assay showed that the proliferation of ASMCs was related with TGF-β1 concentration to some extent,while its significant effect was the concentration of 10μg/L(P < 0.05).With the treatment of PD98059,the proliferation of ASMCs decreased significantly(P < 0.05).After treatment of ASMC with TGF-β1 and β-CD,the expression of caveolin-1 in ASMC decreased significantly,while the expression of p-ERK1/2 increased.Conclusion Caveolin-1 suppressed the effect of TGF-β1-induced ASMCs proliferation of asthmatic rats,which may be involved with ERK1/2 pathway.
