电纺合成聚偏二氟乙烯补片体外细胞毒性及细胞相容性研究

摘　　要：目的评价人工合成聚偏二氟乙烯（polyvinylidene fluoride,PVDF）盆底补片的细胞毒性及细胞相容性。方法将小鼠成纤维细胞（L929细胞）与A组（空白对照组）、B组（乳胶手套组）、C组（聚丙烯组）和D组（PVDF组）浸提液共培养24、48、72h。采用倒置显微镜观察细胞形态学变化,四唑盐MTS比色法检测细胞增殖率（FGR）并进行毒性分级,流式细胞仪检测72h细胞凋亡情况。将L929细胞种植于PVDF材料上,扫描电镜观察第1、3、7、14天时细胞生长情况。结果 A组、C组和D组24、48、72h细胞生长良好,B组见大量死细胞,细胞层几乎完全破坏;B组、C组和D组24、48、72h细胞毒性分别为4级、1级和0-1级;A组、C组和D组72h存活细胞比例分别为（92.43±1.24）%、（92.23±0.64）%和（92.80±0.89）%,三组比较,差异无统计学意义（P〉0.05）;扫描电镜提示,细胞在PVDF材料上可发生黏附增殖。结论PVDF材料补片对L929细胞不具有细胞毒性,并具有良好的体外细胞相容性。Objective To study the cytotoxicity and compatibility of polyvinylidene fluoride （PVDF） materi- al mesh. Methods Mouse fibroblast cell cultivated by four groups leaching solution for 24 h, 48 h, 72 h, which were group A （ blank control group ） , group B （latex gloves）, group C （polypropylene） and group D （PVDF）. Morphologic change was observed by inverted microscope and relative growth rate was detected by MTS assay . And flow cytometry on 72 h to observe cell apoptosis. The growth of cell was observed by scanning electron micros- copy （SEM） on the first day, the third day and on the seventh day and the 14th day. Results L929 cells grew well in group A, group C and group D on 24 h, 48 h and 72 h. The cell toxicity grade of Group B, group C and group D were level 4, 1, 0 to 1 compared to group A. Ratio of normal cells of group A, group C, group D were （92.43±1.24） %, （92. 80±0. 89）% and （92.23±0.64）% by flow cytometry, which was no statistically significant differ- ence. The SEM observed that L929 cell adhere and grow on PVDF mesh. Conclusion The PVDF material mesh has no cytotoxicity and has good cell compatibility to L929 cell.
