摘 要:目的:对大鼠坐骨神经电损伤后疼痛行为进行定量检测,探索周围神经电损伤后出现的感觉功能障碍。方法:成年雄性SD大鼠56只,随机分成假手术组、电损伤65 V、75 V、100 V、125 V、150 V和坐骨神经慢性压榨(CCI)组,共7组,每组8只大鼠。分别于术后1、2、4周对大鼠后足进行热痛敏和机械痛敏定量检测。结果:和对照组相比65 V和75 V组表现出明显的机械缩足反射阈值减小,125 V组1周时机械缩足反射阈值减小,而2周和4周时则明显增大。150 V组1周机械缩足反射阈值无明显变化,2周和4周时明显增大。100 V、125 V和150 V均表现为热刺激缩足反射潜伏期延长。CCI组不论机械还是热刺激,其缩足反射阈值均减小。结论:坐骨神经电损伤可引起大鼠机械性痛阈值和热痛阈值的改变,其中较低电压损伤组大鼠机械性痛阈值减小,而较高电压组大鼠机械刺激阈值和热刺激阈值反而增高,说明较高电压可能导致坐骨神经的感觉传导功能完全受损。Objective: To perform quantitative analysis of the pain behavior after sciatic nerve electrical injury in rats and to investigate the resulting sensory dysfunction. Methods:56 adult male SD rats were randomly divided into seven groups:sham group,65 V,75 V,100 V,125 V and 150 V electrical injury groups, and chronic constriction injury( CCI), each group contained 8 rats. After 1,2 and 4 weeks, respectively, quantitative detection of thermal hyperalgesia and mechanical hyperalgesia were conducted. Results: Compared with the control group, 65 V and 75 V groups showed lowered mechanical threshold for paw withdrawal. At the first week 125 V paw withdrawal mechanical threshold decreased significantly,but increased at 2 weeks and 4 weeks. Paw withdrawal mechanical threshold in 150 V group at the first week had no significant changes, but significantly increased at two and four weeks. The latency of paw withdrawal to thermal stimuli in 100 V, 125 V and 150 V were prolonged. Both of the paw withdrawal mechanical threshold and paw withdrawal thermal latency decreased in CCI group. Conclusion:Sciatic nerve electrical injury altered the threshold of mechanical hyperalgesia and thermal hyperalgesia. Lower voltage stimulation decreased the mechanical threshold, and the higher voltage stimulation can increase both the mechanical and the thermal stimulation threshold. These results indicated that higher electric voltage may impair the sensory propagation of sciatic nerve after electrical injury.
