摘  要:目的探讨甲状腺素联合多奈哌齐对成年期甲状腺功能减退模型SD大鼠的海马乙酰胆碱酯酶活性及乙酰胆碱含量和突触结合蛋白(Syt-1)表达的影响。方法将成年SD大鼠随机分为正常对照组、甲状腺功能减退组(甲减组)、单独甲状腺素治疗组和甲状腺素联合多奈哌齐治疗组(联合治疗组),每组10只。采用色度法测定各组大鼠海马乙酰胆碱酯酶活性和乙酰胆碱含量,并应用免疫组织化学法检测大鼠海马不同亚层Syt-1表达。结果与正常对照组相比,甲减组大鼠海马CA1、CA3和齿状回区乙酰胆碱酯酶活性、乙酰胆碱含量及Syt-1表达显著降低(P<0.05)。单独甲状腺素治疗组大鼠海马乙酰胆碱酯酶活性和乙酰胆碱含量恢复正常,但Syt-1蛋白表达未恢复。联合治疗组大鼠海马乙酰胆碱含量和Syt-1表达均恢复至正常。结论甲减可引起脑内乙酰胆碱酯酶活性、乙酰胆碱含量及Syt-1表达下降,甲状腺激素单独治疗对上述损伤有改善作用,但联合多奈哌齐治疗效果可能更好。Objective To investigate the effects of thyroxine and donepezil treatment on the hypothyroidism-induced alterations of acetylcholine (ACh) content and acetycholinesterase (ACHE) activity and synaptotagmin-1 (Syt-1) expression in the hippocampus of adult rats.Methods Adult male SD rats were randomly divided into three groups:blank control group,hypothyroidism group,thyroxine treatment group,thyroxine and donepezil treatment group.Concentration of ACh and the activity of AChE were determined colorimetrically,and protein level of Syt-1 was determined by immunohistochemistry.Results Compaired to the controls,the hypothyroidism rats had significant decrements of Ach levels,AChE activity and Syt-1 expression in the hippocampus (P < 0.05).Thyroxine,ACh levels and AChE activity were restored by thyroxine administration,while expression of Syt-1 did not reach the control level.Co-administration of thyroxine and donepezil led to normalization of thyroxine,ACh levels as well as the Syt-1 expression.Conclusion Hypothyroidism can lead to the decline of ACh levels,AChE activity and Syt-1 expression in the hippocampus,and thyroid replacement alone may reduce some of these damages,but thyroid combined with donepezil treatment can be better in the hypothyroidism adult rats.
