摘 要:目的探讨骨髓间充质干细胞(BMSCs)和胚胎干细胞(ESCs)因子对迟发型超敏反应(DTH)的影响。方法分离、培养并传代小鼠BMSCs和ESCs,获取BMSCs因子和ESCs因子。小鼠随机分为正常组(A组)、模型组(B组)、培养基组(c组)、地塞米松(DEX)组(D组)、BMSCs因子组(E组)和ESCs因子组(F组)6组,每组10只,应用5g/L二硝基氟苯腹部涂抹致敏、耳廓激发的方法制备小鼠变应性接触性皮炎(ACD)模型(A组不处理),A、B组尾静脉注射生理盐水,C、D、E、F组分别注射DMEM、DEX、BMSCs因子、ESCs因子,比较各组耳肿胀度、胸腺指数和脾指数,光镜下观察耳廓苏木精一伊红染色后组织学变化。结果B组耳肿胀度高于A组,差异有显著性(F=153.080,q=27.68,P<0.01);D组、E组和F组小鼠耳廓肿胀度均低于B组(q=8.130~20.440,P<O.01),脾指数和胸腺指数亦低于B组(F=621.352、409.533,q=9.579~38.030,P<O.01);E组和F组的脾指数及胸腺指数均高于D组(q=4.089~15.969,P<0.05),E组耳肿胀度、脾指数及胸腺指数均高于F组(q=4.860~11.487,P<O.05)。B组和C组以上指标比较差异均无显著性(P>O.05)。镜下观察,D组、E组和F组炎性细胞浸润均较B组减少。结论BMSCs因子和ESCs因子均可抑制DTH,但ESCs因子抑制作用优于BMSCs因子。Objective To explore the influence of cytokines secreted by bone mesenchymal stem cells (BMSCs) and embryonic stem cells (ESCs) on delayed type hypersensitivity (DTH). Methods BMSCs and ESCs cytokines were obtained by iso lating, developing and extending mouse BMSCs and ESCs. The mice were randomized to normal group (group A), model group (group B), medium group (group C), dexamethasone group (group D), BMSCs cytokines group (group E) and ESCs eytokines group (group F), with 10 mice in each group. Mouse model of allergic contact dermatitis(ACD) was created by sensitization through painting 5 g/L dinitrofluorobenzene (DNFB) on abdomen of mice and provocation on ears (group A was excepted). The mice in groups A and B were given intravenous normal saline, and those in groups C, D, E, and F were offered intravenous DMEM, DEX, BMSCs and ESCs, respectively. The degree of swelling of ear, thymus and spleen indexes were compared among each group and auricle histological diversities after HE staining were observed under light microscopy. Results The degree of swelling of ear in group B was more severe than group A, the difference was significant (F=153.080,q:27.68,P〈O.Oa), that in groups D, E and F was milder than in group B (q=8.130 20.440,P〈0.01), the thymus and spleen indexes were also lower than that in group B (F=621.352,409.533;q= 9.579-38.030;P〈0.01). Thymus and spleen indexes in groups E and F were higher than that in group D (q=4.089-15.969,P〈0.05). The extent of swelling of ear and spleen and thymus indexes in group E were higher than that in group F (q = 4.860-11.487, P〈0.05). The differences of the above indicators between groups B and C were not significant (P〉0.05). Microscopic examination indicated that inflammatory cell infiltration was less in groups D, E and F versus group B. Conclusion Both BMSCs and ESCs can inhibit DTH, but the former surpass the action of the latter.
