益肾祛淋汤剂对糖尿病肾病大鼠足细胞的保护作用

摘　　要：目的探讨益肾祛淋汤剂对糖尿病肾病(DN)大鼠足细胞的保护作用。方法 60只雄性大鼠随机均分成实验组(48只)和正常对照组(N组,12只)。实验组腹腔注射链脲菌素制备糖尿病模型后随机分为糖尿病组(D组)、中药方剂治疗大剂量(DA组)、小剂量(X组)和贝那普利(A组)治疗组。分别于4周及8周处死大鼠,检测相关指标。结果 (1)与N组比较,D组出现血糖、甘油三酯、胆固醇、肌酐、肾重指数和24-h尿蛋白排泄量增高(P<0.01),肌酐清除率下降(P<0.01),肾组织内podocin蛋白表达下调(P<0.01)。(2)与D组比较,DA组、X组和A组肾重指数、24-h尿蛋白定量和肌酐降低(P<0.01)。(3)DA组肾组织内podocin蛋白表达高于A组和X组(P<0.05)。结论益肾祛淋汤剂对DN大鼠有明显的治疗作用;其对肾脏足细胞的保护作用机制可能与上调肾组织内podocin蛋白表达有关。Objective To investigate the protective effect of Yishen Qulin decoction（YQD） on the podocytes of rats with diabetic nephropathy（DN）. Methods Sixty male rats were randomly divided into groups of blank control（N, 12 rats） and DN（48 rats）. DN models in group DN were established by intraperitoneal injection of streptozotocin bacteria, which were equally allocated into 4 groups of D （model control）, DA （treated with large dose of YQD）, X （treated with small dose of YQD） and A（treated with benazepril）. The rats were sacrificed in the 4th and 8th weeks for detecting the DN-related indicators. Results Compared with group N, blood glucose, triglyceride, cholesterol, creatinine（Cr）, relative kidney weight and 24-hour urinary protein excretion were significantly increased,Cr clearance rate and podocin protein expression in renal tissues were decreased in group D （P〈0. 01）. Compared with group D, the relative kidney weight, 24-hour urinary protein quantity and Cr were decreased in groups of DA, X and A（P〈0. 01）. The expression of podocin protein in the kidney was higher in group DA than that in groups of A and X（P〈0. 05）. Conclusion YQD has obvious therapeutic effect on DN rats, which may be related to the upregulation of podocin protein expression in renal tissues.
