高效液相色谱质谱联用法测定人血浆中万古霉素和去甲万古霉素浓度

摘　　要：目的：建立同时测定人血浆中万古霉素和去甲万古霉素浓度的LC-MS/MS方法。方法用25％三氯乙酸-乙腈（2∶1）沉淀蛋白血浆样品，用ZORBAX Extend C18色谱柱，以乙腈-水（均含0.1％甲酸）为流动相梯度洗脱，用电喷雾离子化源，正离子方式，多反应监测（ MRM ）扫描方式进行监测。结果血浆中万古霉素和去甲万古霉素的线性范围均为0.2～100.0μg· mL-1（ r＝0.9978），定量下限为0.2μg· mL -1，平均回收率94.4％～107.2％，日内、日间精密度（ RSD）均小于15％。结论该方法快速、灵敏、准确，专属性强，重复性好，适用于人血浆中万古霉素和去甲万古霉素浓度的测定。Objective To develop an LC -MS/MS method for simulta-neous determination of vancomycin and norvancomycin concentrations in human plasma.Methods The plasma samples were precipitated by 25%trichloroacetic acid -acetonitrile （ 2∶1 ）.The separation was a-chieved on ZORBAX Extend C 18 column with gradient elution using water and acetonitrile （ 0.1% formic acid in both ）.Detection of the analytes were made using positive ion electrospray ionization （ ESI） in the multi-ple reaction monitoring （ MRM） mode.Results The linear calibration curves of vancomycin and norvancomycin were both in the range of 0.2-100.0 μg· mL-1 （ r=0.997 8 ） , with the lower limit of quantitation at 0.2 μg· mL-1.The average recovery was 94.4%-107.2%, Intra-day and inter-day relative standard deviations were both below 15%.Conclusion This method is rapid , sensitive , accurate , specific , and reliable, suitable for simultaneous determination of vancomycin and nor-vancomycin concentration in human plasma.
