摘 要:目的:建立同时测定人血浆中万古霉素和去甲万古霉素浓度的LC-MS/MS方法。方法用25%三氯乙酸-乙腈(2∶1)沉淀蛋白血浆样品,用ZORBAX Extend C18色谱柱,以乙腈-水(均含0.1%甲酸)为流动相梯度洗脱,用电喷雾离子化源,正离子方式,多反应监测( MRM )扫描方式进行监测。结果血浆中万古霉素和去甲万古霉素的线性范围均为0.2~100.0μg· mL-1( r=0.9978),定量下限为0.2μg· mL -1,平均回收率94.4%~107.2%,日内、日间精密度( RSD)均小于15%。结论该方法快速、灵敏、准确,专属性强,重复性好,适用于人血浆中万古霉素和去甲万古霉素浓度的测定。Objective To develop an LC -MS/MS method for simulta-neous determination of vancomycin and norvancomycin concentrations in human plasma.Methods The plasma samples were precipitated by 25%trichloroacetic acid -acetonitrile ( 2∶1 ).The separation was a-chieved on ZORBAX Extend C 18 column with gradient elution using water and acetonitrile ( 0.1% formic acid in both ).Detection of the analytes were made using positive ion electrospray ionization ( ESI) in the multi-ple reaction monitoring ( MRM) mode.Results The linear calibration curves of vancomycin and norvancomycin were both in the range of 0.2-100.0 μg· mL-1 ( r=0.997 8 ) , with the lower limit of quantitation at 0.2 μg· mL-1.The average recovery was 94.4%-107.2%, Intra-day and inter-day relative standard deviations were both below 15%.Conclusion This method is rapid , sensitive , accurate , specific , and reliable, suitable for simultaneous determination of vancomycin and nor-vancomycin concentration in human plasma.
