摘  要:目的:建立同时测定人血浆中茶碱、多索茶碱、二羟丙茶碱和氨茶碱浓度的LC-MS/MS法。方法人血浆样品用乙腈直接沉淀蛋白后,用ZORBAX SB C18色谱柱,以乙腈-10 mmol· L-1乙酸铵溶液(均含0.1%甲酸)为流动相梯度洗脱,用电喷雾离子化源,正离子方式,多反应监测( MRM)扫描方式进行监测。结果血浆中茶碱、多索茶碱、二羟丙茶碱和氨茶碱的线性范围为0.10~50.00μg· mL-1( r >0.99),定量下限为0.10μg · mL-1;平均回收率93.5%~109.8%;日内、日间精密度( RSD)均小于15%;提取回收率在81.2%~90.8%。结论该方法快速、灵敏、准确,专属性强,重复性好,适用于人血浆中4种茶碱类的药代动力学研究。Objective To develop an LC -MS/MS method for simulta-neous determinations of theophylline , doxofylline diprophylline and amin-ophylline in human plasma.Methods The plasma samples were precipi-tated by acetonitrile.The separation was achieved on ZORBAX SB C 18 col-umn and eluted with linear gradient using acetonitrile -10 mmol · L-1 mammonium acetate , both of which were acidified with 0.1%formicacid.Detection of the analytes were made using positive ion electrospray ioniza -tion ( ESI) in the multiple reaction monitoring ( MRM) mode.Results The linear calibration curve of theophylline , doxofylline , diprophylline and aminophyllinewere obtained concentration range of 0.10 -50.00 μg · mL-1 (r>0.99), with the lower limit of quantitation at 0.10 μg· mL-1.The average recovery was between 93.5% and 109.8%, Intra -day and inter-day relative standard deviations were both below 15%.The recoveries were 81.2%-90.8%.Conclusion This method is rapid, sensitive, accu-rate, specific and reliable, suitable for simultaneous determination of theo-phylline, doxofylline, diprophylline and aminophylline in human plasma.
