摘  要:目的探讨创伤后应激障碍(PTSD)大鼠下丘脑-垂体激素分泌的变化。方法选择8周龄雄性SD大鼠40只,随机分为空白组和模型1d、4d、7d、14d组。运用放射免疫法(Radioimmunoassay,RIA)测量大鼠血浆促肾上腺皮质激素(ACTH)和皮质醇(COR)的含量,以及血清中促甲状腺激素(TSH),四碘甲状腺原氨酸(T4)和三碘甲状腺原氨酸(T3)的含量。结果①各组大鼠血浆ACTH和COR含量的变化:与空白组比较,血浆ACTH含量有升高趋势,其中模型14d组最明显,具有显著差异性,P〈0.01;血浆COR含量也呈现升高趋势,其中模型14d组最明显,具有显著差异性,P〈0.05。②各组大鼠血清TSH、T4和T3含量变化:与空白组比较,模1d组T4含量明显上升,两组比较差异有统计学意义,P〈0.05。模4d组、模7d组,模14d组T4含量呈下降趋势,且模14d组Ta含量降低最明显,与空白组比较,差异有统计学意义,P〈0.01。5组大鼠的TSH、T3水平差异无统计学意义,P〉0.05。结论创伤后应激障碍早期大鼠体内存在下丘脑一垂体分泌调节功能紊乱。Objective To investigate the changes of secretion of neuroendocrine hormones in post-trauma stress disorder (PTSD). Methods Forty SD rats were randomly divided into 5 equal grous: blank control group, and 1 d, 4 d, 7 d, and 14 d PTSD model groups. The model groups were confined in ?? for 2 h, forced to swim for 15 rain, underwent ether anesthesia, and then fed conventionally. 1 d, 4 d, 7 d, and 14 d later respectively, they underwent collection of serum from ?? vein to detect the levels of thyroid-stimulating hormone (TSH), triiodothyronine (T3), and tetraiodothyronine (T4) by radioimmunoassay (RIA); and underwent laporotomy to collect plasma from abdominal aorta to detect the levels of adrenocorticotrophic hormoine (ACTH) and cortisol (COR). Results (1) Both the ACTH level and COR level of the model groups had a tendency to increase, and the ACTH and COR levels of the model 1 d group were both significantly higher than those of the control group (both P 〈0.01). (2) The T4 level of the 1 d group was significantly higher than that of the control group (P 〈0.05), the T4 levels of the 4 d, 7 d, and 14 d groups showed a tendency of decrease, and the T4 level of the 14 d group was significantly lower than that of the control group (P 〈0.01). There were no significant differences in the TSH and T3 levels among these 5 groups (all P 〈0.01). Conclusion There exists hypothalamus-pituitary-thyroid secretion dysfunction early in PTSD.
