摘 要:目的观察评价人永生化尿路上皮细胞系(immortalized human bladder urothelium cell line)SVHUC-1在胶原支架中的生长情况,为进一步体内组织修复研究提供依据。方法制备疏松多孔的胶原支架材料,SV-HUC-1传代扩增后,接种在胶原支架上,体外培养7 d后取材,观察SV-HUC-1和胶原的复合情况。结果 SV-HUC-1呈"多角形"、"铺路石"样,以克隆团形式生长。HE染色显示胶原支架上的SV-HUC-1经过体外培养7 d后即可形成沿支架表面排列的复层上皮结构,大量SV-HUC-1渗透到胶原的空隙内。扫描电镜显示细胞材料复合良好。结论 SV-HUC-1能在胶原支架上较好的生长,胶原可以作为理想的支架材料,进一步用于组织工程膀胱修复的研究。[ Objective] To observe the growth of immortalized human bladder urothelium cell line (SV- HUC-1) on collagen biomaterials cultured in vitro. [Methods] Porous collagen scaffolds was prepared. After in vitro culture and amplification, the SV-HUC-1 were seeded onto the collagen scaffolds. After cultured for 7 days, the cellscaffolds composites were observed. [Results] SV-HUC-1 were polygon and paving stonelike, and they could form the clone groups. After 1 week of in vitro culture, the SV-HUC-1 formed a muftilayer structure on the surface of the scaffolds, and a great deal of cells grew into the scaffolds. Scanning electron microscopy revealed that cell and scaffolds composited well. [Conclusion] Collagen scaffolds have a good biocompatibility with SV-HUC-1, which can be used as perfect scaffolds in tissue defect reconstruction with the application of tissue engineering.
