摘 要:[目的]揭示肌肉生长抑制素与氨基酸营养的关系,为阐明鹅肌肉生长发育的调控机理提供依据.[方法]利用分子生物学技术克隆吉林白鹅MSTN基因,通过饲喂4种不同的日粮氨基酸模型研究不同日粮氨基酸模型对1~ 28日龄和29~56日龄的吉林白鹅MSTN基因表达量的影响.[结果]28d,A组胸肌MSTN mRNA表达量极显著高于其他3组(P<0.01),A组和C组腿肌MSTN mRNA表达量显著高于B和D组(P<0.05),在B组日粮氨基酸模型下胸肌和腿肌的MSTN mRNA表达量与胸肌率和腿肌率呈极显著负相关.56 d,A组和D组胸肌MSTN mRNA表达量显著高于B组和C组(P<0.05),D组腿肌MSTN mRNA表达量显著高于A、B、C组(P<0.05),在C组日粮氨基酸模型下胸肌和腿肌MSTNmRNA表达量与胸肌率和腿肌率呈极显著负相关.[结论]28d,日粮氨基酸模型B能够抑制MSTN基因的表达,而56 d日粮氨基酸模型C能够抑制MSTN基因的表达,并且MSTN基因表达量与胸肌率和腿肌率之间存在极显著负相关.[Objective] The study aimed to reveal the relationship between MSTN and amino acid nutrition and provide basis for elucidating the regulation mechanism of the growth and development of muscles.[Method] MSTN gene of Jilin White Goose was cloned by using molecular biology techniques.The geese were fed with four different amino acid dietary models to study the effects of different amino acid dietary models on the expression amount of MSTN gene of 1-28 day-old and 29-56 day-old Jilin White Goose.[Result] On the 28-day-old,the expression amount of MSTN mRNA in chest muscle in group A was extremely significantly higher than that in other three groups (P < 0.01).And the expression amount of MSTN mRNA in leg muscle in group A and C were significantly higher than that in group B and D (P < 0.05).In group B,the expression amount of MSTN mRNA in chest muscle and leg muscle showed extremely significantly negative correlation with the chest muscle rate and leg muscle rate.On the 56-day-old,the expression amount of MSTN mRNA in breast muscle in group A and D were significantly higher than that in group B and C (P < 0.05).The expression amount of MSTN mRNA in leg muscle in group D was significantly higher than that in other three groups (P < O.05).In group C,the expression amount of MSTN nRNA in chest muscle and leg muscle showed extremely significantly negative correlation with the chest muscle rate and leg muscle rate.[Conclusion] On the 28-day-old,amino acid dietary model B could suppress the expression of MSTN gene.On the 56-day-old,amino acid dietary model C could inhibit the expression of MSTN and the expression amount of MSTN showed significantly negatively correlation with the chest muscle rate and leg muscle rate.
