速康解毒口服液对小白鼠的急性毒性试验和亚慢性毒性试验

摘　　要：通过小白鼠急性毒性试验和亚慢性毒性试验评价"速康解毒口服液"治疗动物疯草中毒的安全性。选取40只小白鼠,随机分为5组,按200、400、800、1 600mg/kg剂量1次性灌胃给药,观察中毒症状,记录死亡数并计算半数致死量(LD50);另取20只小白鼠,随机分为2组,给药组以最大质量浓度(1.0g/mL)、最大容积(0.8mL)1次性灌胃受试药物,对照组用等体积生理盐水,给药后连续观察7d,测定最大给药量;再取80只小白鼠,随机分成药物处理高、中、低剂量组和对照组,药物组按3 200、1 600和800mg/kg剂量灌胃给药,连续30d,对照组用9g/L生理盐水,称量小白鼠体质量、检测血常规指标、血液生化指标、脏器系数及组织病理变化情况。结果表明,急性毒性试验各剂量组均无小白鼠死亡,无法计算LD50,经口给药的最大剂量为40.0g/kg,表明该受试物无毒;亚慢性毒性试验中,药物处理组小白鼠血常规、血液生化指标和脏器系数与对照组相比无显著差异(P>0.05),观察实质器官无异常病变。提示临床合理使用"速康解毒口服液"不会对靶动物产生毒性作用。To investigate the safety of＂Sukang detoxification oral liquid＂for treatment and prevention of the mastitis in animal,the acute and sub-acute toxicity was tested in mice.＂Sukang detoxification oral liquid＂was extracted with water and concentrated 1.0g/mL solution.In acute toxicity test,40 mice were randomly divided into five groups,the animals were administered drug by intragastric infusion at single dose of 200,400,800,and 1 600mg/kg respectively.Poisoning typical symptom and death cases were observed and the median lethal dose（LD50）were calculated.In order to estimate themaximum dosage,20 mice were randomly divided into the treatment group and control group,＂Sukang detoxification oral liquid＂was administered intragastricsally at maximum concentration（1.0g/mL）and maximum capacity（0.8mL）with single dose in the treatment group,9g/L saline was used in control group.After being administered,the mice were observed for 7days.In sub-acute toxicity test,80 mice were randomly divided into three drug-treated groups（high,middle and low dose）and one control group,drug was administered orally at dose of 3 200,1 600,and 800mg/kg respectively for 30 days.In control group,mice were administered 9g/L NaCl.Routine blood test indexes,blood biochemical parameters,organ coefficient and histopathology were detected.The results showed there was not any poisoning and death cases in each group,there was no LD50 in this compound preparation.The maximum oral dosage of＂Su kang detoxification oral liquid＂was over 40.0g/kg in mice.These data indicated that the drug was actually non-toxin.There were not significantly different in routine blood test,blood biochemical indexes,organ coefficient and vis parenchymal organs disease between three drug treated groups and control group.All of these results suggested that＂Sukang detoxification oral liquid＂was safety for animals,which were in mastitis under the treatment in clinic.
