摘  要:优化隐丹参酮脂质体的最佳制备工艺条件。采用溶剂分散法制备隐丹参酮脂质体,以包封率为指标,以药脂比、膜材比、反应温度和反应时间为因素,采用L9(34)正交试验设计对制备条件进行优选,透析法测定隐丹参酮脂质体的包封率,透射电镜和激光粒度仪测定其形态和粒径。溶剂分散法制备隐丹参酮脂质体的最佳工艺为:药脂质量比1∶20,膜材质量比4∶1,反应温度30℃,反应时间6min;平均包封率为(87.31±1.45)%;粒径集中分布在(176±5.65)nm,且隐丹参酮脂质体形态和粒径均匀,重现性好。To optimize the preparation procedure of cryptotanshinone liposome.Cryptotanshinone liposome was prepared by solvent distribution method.L9(34)orthogonal test was used to optimize the preparation procedure designed with four factors of lecithin to drug ratio,lecithin to cholesterol ratio,reaction temperature and reaction time,and index of encapsulation efficiency.The encapsulation efficiency of Cryptotanshinone liposome was assayed by dialysis method,the shape and particle size were observed by transmission electron microscope and Malvern laser particle size analyzer.The optimized preparation conditions were as follows:the ratio of lecithin to drug was 1∶20,the ratio of lecithin to cholesterol was 4∶1,reaction temperature was 30℃and reaction time was 6min,the mean encapsulation efficiency was(87.31±1.45)%.The particle size was distributed in(176±5.65)nm.Conclusion:the Cryptotanshinone liposome prepared in the optimized conditions has following advantages:high encapsulation efficiency and drug-loading rate,uniform shape and particle size,and good reproduction quality.
