摘  要:目的 探讨高氯酸钠(NaClO4)处理制备兔气管支架的最适宜浓度.方法 健康新西兰兔30只,随机分5组(n=6).以新鲜气管为对照A组,按NaClO4浓度分别为3%、5%、6%、7%为实验B、C、D、E组.各组气管基质经形态学观察和生物力学性能检测比较.结果 组织学染色显示A组气管有大量黏膜上皮细胞,C组气管黏膜上皮细胞去除;电镜扫描可见A组气管上皮细胞的纤毛,实验B组气管基质内表面毛糙欠光滑,C组气管基底膜完整无损坏,而D、E组气管则出现不同程度的裂痕;比较于原生对照组,C组的细胞核去除显著,DNA下降差异有统计学意义(P<0.05),而胶原蛋白、弹力纤维和网状纤维相对保留,基质内的软骨细胞活力良好,糖胺聚糖含量降低差异无统计学意义(P>0.05);生物力学分析:随着NaClO4浓度升高,最大拉伸力、拉伸应变在断裂和弹性模量逐步降低(P<0.05).结论 5%浓度NaClO4处理的兔气管支架保留了较完整的细胞外基质(ECM)结构和理想的脱细胞效果,适合用于构建组织工程同种异体气管.Objective To investigate the optimum concentration of NaClO4 treatment in order to prepare tracheal scaffold.Methods Thirty healthy New Zealand rabbits were randomly divided into 5 groups (n =6),the tracheas of group A was treated with nothing as control group,the tracheas of group B,C,D and E were treated respectively with different concentrations of NaClO4(3%,5%,6%,7%).We obtained the histologic appearance via optical microscope,the ultrastructure was observed through scanning electron microscope (SEM) and survey the structural integrity and biomechanical property of tracheal matrix.Results Histology staining showed vividly lots of epithelial cells in group A,while removed clearly in group C.SEM emerged distinctly the cilium of epithelial cells in group A.The internal surface of group B were still rough,and became smooth in group C.However the matrix in group D and E fractured more or less.Compared to the native group,the number of cell nucleus in group C were obviously decreased,the decline of DNA was statistical significance (P < 0.05).There were not significantly elimination of the collagen,elastic fibers and reticular fibers in group C,as well as the contents of glycosaminoglycan (GAG) (P > 0.05).We found the chondrocyte viability reserved well,but exhibited a significantly lower value of maximum tensile strength,tensile strain at break and elastic modulus with the increased concentration of NaClO4 (P < 0.05).Conclusion Decellularized rabbit tracheal scaffold acquired via 5 % NaClO4 concentration treatment showed its structural integrity,antigen cell removal and a fine biomechanical property,which would be suitable for the application in tissue engineering tracheal allograft.
