摘 要:在对PCR反应体系进行正交试验优化的基础上,采用2 878对SSR分子标记分析以桑树品种伦教109作为父本,珍珠白和粤武2号分别作为母本的2对杂交组合亲本间的多态性。结果表明,桑树杂交组合珍珠白×伦教109亲本间的多态性为69%,粤武2号×伦教109亲本间的多态性为54%。选用珍珠白×伦教109的F1群体进行SSR分子标记遗传连锁分析,对亲本的遗传背景分析发现:在母本珍珠白中,相引相分子标记数目为58对,相斥相分子标记数目为70对,符合1∶1分离比;而在父本伦教109中则没有检测到相斥相分子标记对。据此推测桑树品种珍珠白可能为异源四倍体,而伦教109可能为同源四倍体。The polymorphisms between parents in two mulberry hybrid combinations prepared using Lunjiao 109 as paternal stock and Zhenzhubai and Yuewu 2 as maternal stock respectively were analyzed using 2 878 pairs of SSR markers based on PCR reaction systems optimized by orthogonal test.The results revealed that the polymorphisms between Zhenzhubai × Lunjiao 109 and Yuewu 2 × Lunjiao 109 were 69% and 54%,respectively.SSR molecular marker linkage analysis was performed to F1 population of Zhenzhubai × Lunjiao 109 for analyzing the heredity background of parental stocks.It was found that the number of SSR markers of maternal stock Zhenzhubai linked in coupling and repulsion phase was 58 and 70,respectively,with a ratio close to 1∶ 1.However,no molecular marker pair in repulsion phase was detected in Lunjiao 109.These data suggest that mulberry cultivar Zhenzhubai is an allotetraploid,whereas Lunjiao 109 is an autotetraploid.
