摘  要:热激蛋白(heat shock protein,HSP)在保护细胞免受损伤或降低受损程度等方面发挥重要作用。BmsHSP27.4(GenBank登录号:AK382044)是从家蚕幼虫脂肪体中克隆的一个热激蛋白基因,蛋白质同源性分析发现BmsHSP27.4区别于以往发现的小热激蛋白,单独处于一个分支。以耐高温家蚕品种7532和对高温较敏感的普通家蚕品种皓月为材料,运用半定量RT-PCR和实时荧光定量PCR技术检测高温(34℃)条件下,BmsHSP27.4基因在不同家蚕品种以及同一家蚕品种不同性别5龄幼虫的中肠、丝腺和脂肪体组织中的表达水平。结果显示:高温处理后该基因表达水平在2个家蚕品种雌雄幼虫的3个组织中均呈快速上升的趋势,说明该基因是高温敏感基因;耐高温品种7532雄蚕脂肪体和中肠组织中该基因的表达水平高于雌蚕。该基因的表达可能与家蚕的耐热性有关。Heat shock protein(HSP) plays important roles in protecting or reducing the cells from damage.BmsHSP27.4(GenBank accession No.AK382044) is a heat shock protein gene cloned from fat body of the5 th instar silkworm(Bombyx mori).Homologous analysis showed that BmsHSP27.4 belongs to an evolutionary clade different with other known small heat shock proteins.In this study,high temperature resistant silkworm strain 7532 and normal silkworm strain Haoyue were used as material to determine expression level of BmsHSP27.4 in midgut,silk gland,and fat body from the 5th instar silkworm larvae of different varieties and different sexes under treatment of high temperature(34 ℃) by semi-quantitative RTPCR and qRT-PCR.The results showed that BmsHSP27.4 expression increased rapidly in the three tissues of male and female larvae of the two silkworm strains,indicating that BmsHSP27.4 was a sensitive gene to high temperature.In silkworm strain 7532,BmsHSP27.4 displayed higher expression level in fat body and midgut from male larvae than that from female larvae.It is concluded that expression of this gene is associated with high temperature resistance of silkworm.
