摘 要:害虫对杀虫剂的抗性遗传特性是农林业害虫防治体系中制定抗性治理对策的重要依据。以鳞翅目昆虫家蚕作为模型,研究害虫对微生物杀虫剂苏云金芽孢杆菌(Bt)毒素的抗性遗传特性。在调查100个家蚕品系对Bt毒素Cry1Ac的抵抗能力的基础上,运用经典的杂交和回交方法,以家蚕抗性品系ACR1(R,LC50>375 mg/L)和敏感品系Nistari(S,LC50=37.5 mg/L)为亲本建立正反交F1、自交F2及回交BC1群体,进一步分析家蚕对Bt毒素Cry1Ac的抗性遗传模式。通过对供试家蚕群体2龄幼虫添食Bt毒素Cry1Ac后24 h的死亡与存活表型分离数据进行统计分析,最终确定家蚕抗性品系ACR1对Bt毒素Cry1Ac的抗性由隐性单基因控制。The inheritance character of pest-insect’s resistance to insecticide is the important basis for formulating resistance management strategy in controlling agricultural and forestry pests.Using silkworm(Bombyx mori) as a model of lepidopteran insect,we investigated the inheritance character of pest-insect’s resistance to Bacillus thuringiensis(Bt) toxin,which is a kind of microbial insecticide.The resistivity of 100 silkworm strains against Bt toxin Cry1 Ac was investigated.Then,classical crossing and back-crossing methods were employed to establish reciprocal crossing F1,self-crossing F2,and back-crossing BC1 populations from resistant silkworm strain ACR1(R,LC50〉 375 mg /L) and sensitive silkworm strain Nistari(S,LC50= 37.5 mg /L) for studying the inheritance pattern of silkworm resistance to Bt toxin Cry1 Ac.Through statistical analysis on segregation data of the death and survival phenotypes of the 2nd instar silkworm larvae at 24 h after treatment of Bt toxin Cry1 Ac,we finally confirmed that the resistance of the resistant silkworm strain ACR1 to Bt toxin Cry1 Ac is controlled by a recessive single gene.
