摘  要:目的探讨早期嗅觉剥夺对SD大鼠海马齿状回及其所含GABAA受体δ亚基表达的影响。方法将3日龄与8日龄SD乳鼠各36只均随机分为空白对照组、实验组、假手术组,共6组。其中通过三氯甲烷滴注法对实验组乳鼠的嗅粘膜进行破坏;假手术组用生理盐水替代;空白对照组不给予任何处理。各组在4周龄时断头取脑组织进行尼氏染色观察;免疫组织化学相对定量分析海马齿状回中突触外GABAA受体δ亚基相关蛋白的表达;实时荧光定量核酸扩增(q-Pcr)定量分析海马中突触外GABAA受体δ亚基mRNA的表达。结果尼氏染色显示3d实验组乳鼠海马齿状回神经元细胞排列紊乱,细胞间距加大,胞质内尼氏体减少,齿状回颗粒细胞边界不齐、间距增宽、排列松散紊乱。3d实验组乳鼠与8d组比较,海马齿状回神经元数目差异均无统计学意义(P>0.05)。而3d实验组乳鼠中海马齿状回突触外GABAA受体δ亚基相关蛋白和mRNA的表达均下降,分别与3d假手术组和空白对照组比较差异有统计学意义(P<0.01)。在8d实验组、8d假手术组和空白对照组之间结果进行两两比较中,差异均无统计学意义(P>0.05)。结论 3d实验组SD鼠海马中突触外GABAA受体δ亚基相关蛋白及mRNA表达显著减少,表明早期嗅觉剥夺对海马齿状回及其所含突触外GABAA受体δ亚基表达的降低有影响。Objective To explore impact of early olfactory deprivation on hippocampus and extrasynaptic GABAA receptor delta subunits in dentate gyrus of SD rats. Methods 36 3 - day - old - rats and 36 8 - day - old - rats were divided randomly into 3 groups : blank control group, experimental group and sham oper- ation group. Experimental group: 3 - day - old experimental group and 8 - day - old experimental group of rats established by chloroform fixed point injection to damage the nasal mucosa. Sham operation group: the same method of the experimental group by dropping physiological saline instead of dropping chloroform. Blank control group : without any treatment. Each group was beheaded at 4 weeks ages, and took the brain organiza- tion to conduct the Nissl Staining, delta subunit immunohistochemistry and q -PCR research in hippocampus. Results The findings of Nissl staining showed that neurons in the hippocampal DG of 3 -day -old experi- mental group arranged in disorder, cell spacing increased, cytoplasmic Nissal bodies decreased; DG cell boundary irregularity, cell space increased, cells arranged in disorder. There was no significant difference of the number of hippocampal neurons among blank control group, experimental group and sham operation group whatever in 3 - day - old - rats and 8 - day - old - rats groups ( P 〉 0.05 ). Related quantitative analysis of immunohistochemistry showed that expression of B subunit of 3 - day - old experimental group represented much less than 3 -day- old sham operation group and blank control group in hippocampus(P 〈0.01 ). The find- ings of q - PCR showed that mRNA of subunit of the hippocampus of 3 - day - old sham operation group and blank control group much more than 3 -day -old experimental group( P 〈 0.01 ). But there was no significant difference among blank control group, experimental group and sham operation group at 8 - day - old rats what- ever in immunohistoehemistry and q - PCR research(P 〉 0. 05 ).
