摘  要:扬稻6号是目前两系杂交籼稻生产中应用面积最大的恢复系,通过“实践八号”育种专用卫星搭载扬稻6号进行航天诱变,筛选出一个光温敏雄性核不育系扬稻6号S。以扬稻6号S与井冈旱稻1号杂交并回交的F2及BC2F2为定位群体进行不育基因的遗传分析。结果表明:扬稻6号S不育性受1对隐性不育基因(暂命名为ptgms2—2)调控,该不育基因被定位于第2染色体标记InDel2-4085和1nDel 2-5846之间,物理距离为200kb。对候选基因进行测序分析,发现在开放阅读框内起始密码子下游70~71bp处存在差异,扬稻6号为GC,扬稻6号S为TA,2个碱基突变导致密码子由GCG变为终止密码子TAG。Yangdao 6 was the most widely used restorer line in the indica two-line hybrid rice breeding system. A pho- toperiod-thermo-sensitive genic male sterile line Yangdao 6 S was selected from the space mutation population of Yangdao 6 via breeding satellite Shijian 8. Genetic analysis indicated that the sterility of Yangdao 6 S was controlled by a single recessive gene using the F2 and BC2F2 population derived from a cross of between Yangdao 6 S and Jingganghandao 1. The sterile gene ptgrns2-2 was restricted to a 200 kb region on chromosome 2 between the marker InDel 2-4085and InDel 2-5846. The sequencing results indicated that there were two nucleotide polymorphisms between Yangdao 6 and Yangdao 6 S. The nucleotide G and C mutated into T and A at site 70-71 bp, subsequently, a premature stop codon was created.
