摘 要:目的:研究幼年大鼠单侧输尿管结扎(UUO)致肾间质纤维化模型中,MicroRNA-192(miR-192)、Ⅰ型胶原的表达以及用免疫抑制剂雷公藤多苷(TWP)的干预效果。方法选用4~5周龄幼年SD雄性大鼠54只,按随机数字表法均分为3组,假手术组、UUO模型组及TWP治疗组,分别于治疗满3、7、14 d杀鼠取左肾,采用实时荧光定量聚合酶链反应(PCR)方法对各组miR-192及Ⅰ型胶原mRNA进行定量检测。结果假手术组miR-192、Ⅰ型胶原mRNA仅微量表达,各时间点组内比较差异无统计学意义(P>0.05)。与假手术组相比, UUO模型组miR-192、Ⅰ型胶原mRNA表达增加(P<0.01),且随着梗阻时间的延长,两者的表达逐渐增强,各时间点组内比较差异有统计学意义(P<0.01);与UUO模型组相比,TWP治疗组可以减低miR-192、Ⅰ型胶原mR-NA的表达(P<0.01),但仍高于假手术组(P<0.01)。结论在幼年大鼠单侧输尿管梗阻致肾间质纤维化模型中, miR-192可能起促进作用;雷公藤多苷可有效抑制miR-192、collagenⅠ的表达,改善肾间质纤维化。Objective To investigate the expression of MicroRNA-192(miR-192)and collagenⅠin kidney in-terstitium in young rats with unilateral ureteral obstruction (UUO), and to explore the effects of tripterygium wilfordii polyglycoside (TWP), an immunosuppressive agent on kidney interstitial fibrosis. Methods Fifty-four male SD rats were randomly assigned into sham operation group (n=18), UUO-model group (n=18) and TWP-treatment group (n=18). Mice were sacrificed at days 3, 7 and 14 for biopsy of the left kidney. The mRNA of miR-192 and Collagen I were detected by reverse transcription polymerase chain reaction. Results The expression of miR-192 and CollagenⅠmRNA were minimally detectable in the sham-operation group at all time points (both P〉0.05). Compared with the sham-operation group, the expression of miR-192 and CollagenⅠmRNA were progressively increased with experiment time in the UUO model group (both P〈0.01). Compared with the UUO group (P〈0.01), treatment with TWP led to a significant reduction in the expression of miR-192 and CollagenⅠmRNA, despite the higher levels than control group (P〈0.01). Conclusion miR-192 may play a role in promoting the renal interstitial fibrosis in young rats with UUO. TWP effectively suppresses the synthesis of miR-192 and Collagen I and hinders the progression of renal interstitial fi-brosis.
