摘 要:通过提取大豆DNA,采用浸胚法在不同的浓度和温度下来处理番茄种子,观察后代植株一系列生理指标的变化,通过用紫外分光光度法测定后代植株叶绿素含量,用聚丙烯酰胺凝胶电泳(PAGE)对植株同工酶(SOD)进行分析,发现经处理的番茄种子,后代植株性状产生变异:叶绿素总含量与对照相比出现增加,其中DNA浓度为136.50μg/mL经4℃浸泡避光处理的番茄种子总叶绿素含量变化最大,比对照增加了116%;同工酶(SOD)谱带一致,没有新的谱带出现,但酶活发生了变化。Tomato seeds were treated in different concentrations of soybean DNA and temperature. Changes of a series of physiological indexes of the offsprings were observed. The content of chlorophyll of offsprings was determined by uv spectrophotometry. Polypropylene gel electrophoresis (PAGE)was used to analyze the peroxidase isozyme (SOD) of plants. Results showed that content of total chlorophyll was increased compared with contrast. When the DNA concentration was 136.50μg/mL and tomato seeds were soaked lightly at 4℃, content of total chlorophyll had the biggest changes with increase of 116%. Band of peroxidase isozyme (SOD) was consistent. There was no new band, but the activity of enzyme was changed.
