摘 要:为贵州黑山羊垂体特异性转录因子(Pituitary Specific Transcriptiona Factorl,POU1/Pit-1)与胴体性状相关的分子标记辅助选择提供参考,运用PCR及测序技术对贵州黑山羊POU1F1基因第4内含子进行了多态性检测。结果表明:贵州黑山羊第4内含子存在2个突变位点,即第222位点发生C→T的突变(C222T),产生CC和Cc 2种基因型;第685位点发生G→A的突变(G685A),产生DD和Dd 2种基因型。CC、DD基因型频率均为0.967,C、c基因频率分别为0.983和0.017,D、d基因频率分别为0.983和0.017。等位基因C、D是相应突变位点的优势等位基因,CC、DD基因型是相应突变位点的优势基因型。贵州黑山羊POU1F1基因第4内含子的突变位点达到Hardy-Weinberg平衡。The polymorphism of the fourth intron of POU1F1 gene in Guizhou black goat was detected by PCR and sequencing technique to provide a reference for marker-assisted selection of POU1F1 gene related to carcass trait in Guizhou black goat.There are two mutation loci in the fourth intron of Guizhou black goat,C on 222th nucleotide is mutated to T,which produces CC and Cc genotypes.G on 685 bp is mutated to A,which produces DD and Dd genotypes.The frequency of CC,DD,C,c,D and d genes is 0.967,0.967,0.983,0.017,0.983 and 0.017 respectively.Allelic gene C and D is the dominant allelic gene at the relevant mutation locus separately.CC and DD genotype is the dominant genotype at the relevant mutation locus respectively also.The mutation loci of the fourth intron of POU1F1 gene in Guizhou black goat reaches Hardy-Weinberg equilibrium.
